Strategies to Improve the Lipophilicity of Hydrophilic Macromolecular Drugs
Hydrophilic macromolecular drugs can be successfully lipidized by covalent attachment of lipids, by hydrophobic ion pairing with negatively or positively charged surfactants, and by dry or wet reverse micelle formation. Lipophilicity enhancement of hydrophilic macromolecules has several benefits including stability and bioavailability improvement ...

Distinct sites on ABCA1 control distinct steps required for cellular release of phospholipids1s□
The loss of ABCA1 function leads to Tangier dyslipidemia in humans and to a Tangier-like phenotype in mice, by impairing the transformation of nascent apolipoproteins into mature HDL particles.

Apolipoprotein A‐V Regulation of Apolipoprotein B Secretion
Apolipoprotein ( APO ) A5 is a recently discovered member of the APOA1/C3/A4 gene cluster. Studies with APOA5 transgenic and gene disrupted mice showed that apoA‐V is a key regulator of plasma ...

Structure of apolipoprotein C-IIToronto, a nonfunctional human apolipoprotein. [PDF]
We previously reported a family with apolipoprotein C-II (apoC-II) deficiency characterized by the presence of a mutant apoC-II, apoC-IIToronto. We now report the purification and primary structure of apoC-IIToronto. The sequence of apoC-IIToronto is identical to that of normal apoC-II from residues 1-68.

A high‐throughput in vivo mRNA LNP screening platform is developed and employed to screen a large library of 122 mRNA LNPs in vivo for delivery to immune, stromal, and parenchymal cells, identifying promising LNP candidates. A novel small particle flow cytometry‐based protein adsorption analysis method is utilized to interrogate protein corona ...

In this report, we have identified two apolipoproteins (apo), apoD and apoA-IV, that, together with the previously identified apoA-I and apoE, accumulate in the regenerating peripheral nerve. These four apolipoproteins were identified in regenerating rat sciatic nerves by their molecular weights, their isoelectric points, and their recognition by ...

Helper and ionizable lipids play a crucial role in determining ApoE binding and subsequent liver tropism and LDLR‐mediated uptake. Ionizable lipids primarily govern the LDLR‐independent uptake pathway. This complementary interplay between lipid components ultimately governs LNP delivery performance and therapeutic efficacy in the liver.
