Gossypol, a potent inhibitor of arachidonate 5- and 12-lipoxygenases
Biochimica et Biophysica Acta (BBA) - Lipids and Lipid Metabolism, 1985Gossypol inhibited 5- and 12-lipoxygenases of rat basophilic leukemia (RBL-1) cells with ID50 of 0.3 microM and 0.7 microM, respectively. Nearly two orders of magnitude of higher concentration of gossypol was required to inhibit prostaglandin synthetase. The inhibition was of a non-competitive type with respect to arachidonate.

Arachidonate 12-Lipoxygenase of Human Platelets
1993Thromboxane A2 is well known as a proaggregatory and vasoconstrictive compound [1]. It is produced from arachidonic acid in platelets, and fatty acid cyclooxygenase responsible for the thromboxane synthesis is localized in the microsomal fraction of human platelets [2]. There is another metabolic pathway of arachidonic acid in human platelets.

Inhibition of platelet arachidonic acid 12-lipoxygenase by acetylenic acid compounds
Prostaglandins, 1981Eighteen acetylenic fatty acids were tested as inhibitors of human platelet arachidonic acid 12-lipoxygenase. 4,7,10,13-Eicosatetraynoic (4,7,10,13-ETYA) acid emerged as the most potent compound. Additional experiments have shown that 4,7,10,13-ETYA selectively blocked the 12-lipoxygenase in washed human platelets with lesser activity against the ...

Arachidonate 12-Lipoxygenases: Enzymology and Molecular Biology
1993In contrast to 5-lipoxygenase which is involved in the biosynthesis of bioactive leukotrienes, a general physiological role of 12-lipoxygenase has not yet been established. Although the enzyme was earlier found in platelets as the first mammalian lipoxygenase and has been found in various tissues of many animal species, most investigators have ignored ...

Stimulation by carrageenan of arachidonate 12-lipoxygenase activity in dog gingival tissue
Archives of Oral Biology, 1991At 4 h after injection of carrageenan into the gingiva, the 12-lipoxygenase activity of the gingival homogenate was markedly increased. Activity in the cytosol and microsomal fractions was markedly increased when assessed as the specific activity based on nmol/min/mg of protein, and in the cytosol fraction as the percentage distribution of total ...

Tissue Distribution and Subcellular Localization of Platelet-Type Arachidonate 12-Lipoxygenase
1997Arachidonate 12-lipoxygenase introduces one oxygen molecule regiospecifically and stereoselectively into the carbon 12 of arachidonic acid. Our group has demonstrated the occurrence of two isoforms of 12-lipoxygenase (platelet-type and leukocyte-type) which were distinguished in terms of substrate specificity, the cross-reactivity of their antibodies ...

Docking arachidonic acid into human 5/12-lipoxygenase using ICM
2011 IEEE International Conference on Bioinformatics and Biomedicine Workshops (BIBMW), 2011Lipoxygenases(LOX) are a super gene family of iron-containing enzymes, that catalyze the stereo- and regio-specific formation of fatty acid hydroperoxides from polyunsaturated fatty acids. Mammalian lipoxygenase have been implicated in the pathogenesis of various inflammatory responses.
