Differentiation of Bacteroides ovatus and Bacteroides thetaiotaomicron by means of bacteriophage

Two members of the Bacteroides fragilis group, B. ovatus and B. thetaiotaomicron, are difficult to distinguish by biochemical methods. They are currently identified on the basis of their variable ability to ferment salicin. We studied a method of identification for these two species by using cell lysis by bacteriophages.

The cell-surface antigens of Bacteroides thetaiotaomicron

Three strains of B. thetaiotaomicron of different origin were investigated. Lipopolysaccharides were extracted from the studied strains using a phenol-water method. The best purification of LPS was achieved by digestion with nuclease and subsequent ultracentrifugation. Capsular material (CPS) was obtained from the most heavily encapsulated strain.

Specific growth rates of Bacteroides thetaiotaomicron NCTC 10582 with either glucose, arabinose, mannose, galactose or xylose as sole carbon sources were 0.42/h, 0.10/h, 0.38/h, 0.38/h and 0.16/h respectively, suggesting that hexose metabolism was energetically more efficient than pentose fermentation in this bacterium.

Galactosamine inhibition of protein synthesis in Bacteroides thetaiotaomicron

Galactosamine does not support growth of Bacteroides thetaiotaomicron. Despite this, galactosamine was more effective than utilizable carbohydrates such as glucose in preventing synthesis of the inducible enzymes α-glucosidase and chondroitin lyase. Galactosamine also stopped overall protein synthesis.
