Optimizing photoactivation of PA‐mCherry for optical pooled CRISPR screens
Photoactivatable PA‐mCherry finds widespread use to optically tag individual cells. However, confocal 405 nm UV laser‐scanning (normal scan) is much less efficient than widefield UV illumination, limiting the use of PA‐mCherry on confocal instruments. We remedy this limitation by reporting that rapid and repeated confocal scanning with a low‐intensity,

Bars are structural members that support uni-axial tensile or compressive loadings. Chapter 1 presents elementary examples for bars and bar systems and gives an introduction to inelastic stress analysis. Basic features of inelastic structural responses including creep, relaxation, stress redistribution and others are discussed. In Sect.

In this chapter, we analyze axially loaded structural elements termed bars or trusses. A truss is connected to other elements only through pins which are connections that do not constrain rotations. Trusses are modeled as discrete elements (or springs) because only axial force (traction or compression) and elongation is evaluated.
