Basophils are functionally closely related to mast cells. Both cell types express the high-affinity IgE receptor (FcεRI) and rapidly release preformed mediator from intracellular stores upon IgE-mediated activation. However, in contrast to mast cells basophils finish their maturation in the bone marrow and have a lifespan of only 2-3 days.

We have described a patient with basophilic leukemia secondary to chronic granulocytic leukemia. Symptoms of hyperhistaminemia were aggravated after chemotherapy with hydroxyurea. Antihistamines (H1 receptor antagonists) controlled many of the symptoms related to hyperhistaminemia.

Basophil degranulation control

Then we study an optimal control problem associated to a non-linear compartmental model. This control is associated to an antigen concentration. A 3-step procedure is described in detail for producing 50% basophils in 50% yield from human blood. Leukocytes from dextran sedimented blood or preferably whole blood are sedimented on Hypaque-Ficoll, then on aqueous Hypaque, followed by adherence and elution from a column of glass beads.
