Novel ATR/PARP1 Dual Inhibitors Demonstrate Synergistic Antitumor Efficacy in Triple‐Negative Breast Cancer Models

open access: yesAdvanced Science, EarlyView.
Novel ATR/PARP1 dual inhibitors are developed for the first time. Advanced lead B8 effectively reduces cell viability in vitro and suppresses tumor growth in vivo, with better potency than ATRi and PARPi alone or in combination. Mechanistically, B8 induces apoptosis, arrests the cell cycle, and inhibits cancer cells colony formation, migration, and ...

Some Theory For Practical Classifier Validation [PDF]

open access: yesarXiv, 2015
We compare and contrast two approaches to validating a trained classifier while using all in-sample data for training. One is simultaneous validation over an organized set of hypotheses (SVOOSH), the well-known method that began with VC theory. The other is withhold and gap (WAG).

Inhibition effects of paeonol on mice bearing EMT6 breast cancer through inducing rumor cell apoptosis [PDF]

open access: yes, 2014
Paeonol, a phenolic component from the root bark of Paeonia moutan, has been identified to possess antitumor effects on mice bearing EMT6 breast cancer in our previous studies. However, the underlying mechanisms remain unknown.

Targeted Dual‐Responsive Liposomes Co‐Deliver Jolkinolide B and Ce6 to Synergistically Enhance the Photodynamic/Immunotherapy Efficacy in Gastric Cancer through the PANoptosis Pathway

open access: yesAdvanced Science, EarlyView.
Schematic model of CJP–TiN liposomes for gastric cancer (GC) treatment. Internalizing RGD peptide (iRGD) targets tumor cells, while Chlorin e6 (Ce6) generates reactive oxygen species (ROS). Jolkinolide B (JB) activates the PANoptosis pathway via caspase‐8, enhancing GC cell death and immune response, transforming “cold tumors” into “hot tumors” by ...

Ensemble Validation: Selectivity has a Price, but Variety is Free [PDF]

open access: yesarXiv, 2016
Suppose some classifiers are selected from a set of hypothesis classifiers to form an equally-weighted ensemble that selects a member classifier at random for each input example. Then the ensemble has an error bound consisting of the average error bound for the member classifiers, a term for selectivity that varies from zero (if all hypothesis ...

Targeting the differential addiction to anti-apoptotic BCL-2 family for cancer therapy [PDF]

open access: yes, 2017
BCL-2 family proteins are central regulators of mitochondrial apoptosis and validated anti-cancer targets. Using small cell lung cancer (SCLC) as a model, we demonstrated the presence of differential addiction of cancer cells to anti-apoptotic BCL-2, BCL-

Radiation Exposure Induced Blood–Brain Barrier Injury via Mitochondria‐Mediated Sterile Inflammation

open access: yesAdvanced Science, EarlyView.
This work describes a blood–brain barrier (BBB) microphysiological system (MPS) to explore the responses of BBB to radiation exposure. Following radiation exposure, obvious BBB comprise and brain endothelial injuries are detected. Mechanism study shows radiation induced significant sterile inflammation via mitochondrial dysfunction and cGAS‐STING ...

Computing a Data Dividend [PDF]

open access: yesarXiv, 2019
Quality data is a fundamental contributor to success in statistics and machine learning. If a statistical assessment or machine learning leads to decisions that create value, data contributors may want a share of that value. This paper presents methods to assess the value of individual data samples, and of sets of samples, to apportion value among ...

The simultaneous effect of valproic acid and gamma radiation on telomerase activity and bax and Bcl-2 protein levels in MCF-7 breast cancer cell line [PDF]

open access: yes, 2015
Background: Breast cancer is one of the most prevalent types of cancer. Factors such as ionizing radiation and chemotherapeutic agents can trigger apoptosis and cancer cell death.

Small Extracellular Vesicles Orchestrate Cisplatin‐Induced Ototoxicity: Potential Biomarker and Targets Discovery

open access: yesAdvanced Science, EarlyView.
Cisplatin causes reactive oxygen species accumulation, leading to apoptosis and inflammation in cochlear hair cells. Small extracellular vesicles primarily derived from the damaged hair cells likely contribute to cisplatin‐induced ototoxicity, carrying a variety of microRNAs and proteins.
