Targeting eIF5A combats breast cancer progression by ClpP‐dependent mitochondrial oxidative stress
This study identified the eIF5A‐ClpP axis as a key driver of breast cancer progression. Targeting eIF5A suppresses tumor growth by inducing ClpP‐dependent mitochondrial oxidative stress, revealing a novel therapeutic vulnerability. Following traumatic brain injury, indole‐3‐propionic acid (IPA) directly targets the aryl hydrocarbon receptor on astrocytes in the peri‐lesional cortex; this suppresses interferon regulatory factor 1‐driven transcription of the E3 ubiquitin ligase RFFL (ring finger and FYVE‐like domain containing E3 ubiquitin protein ligase).

This review highlights recent advances in mitochondria‐targeted nanomedicine for cancer therapy. It summarizes key targeting strategies and delineates four principal therapeutic mechanisms: oxidative stress–induced mitochondrial dysfunction, disruption of energy metabolism, direct mitochondrial damage, and induction of immunogenic cell death.
