Stereotyped Subclones Revealed by High‐Density Single‐Cell Lineage Tracing Support Robust Development

Based on a well‐established in vitro directed differentiation model and an integrated analysis of high‐density cell lineage trees (CLTs) and single‐cell transcriptomes, it is demonstrated that many subclones are formed by sub‐CLTs resembling each other in terms of both cell type compositions and topological structures.

NAD+‐Boosters Improve Mitochondria Quality Control In Parkinson's Disease Models Via Mitochondrial UPR

Treatment of MPTP‐incubated cells with NAD+‐boosters increase the UPRmt/mitophagy‐related mitochondria quality control (MQC). Disturbed plasma UPRmt‐mitophagy‐mediated MQC profiles in PD patient samples. NMN inhibits motor deficit and forestalls neuropathology phenotypes of PD mice, which is required the atf4‐medicated UPRmt pathway.

CRISPR/Cas9 screening reveals PTPN2 as essential for ALK+ ALCL survival. PTPN2 regulates TFRC to promote PINK1‐PRKN mitophagy. Oral PTPN2 inhibitor ABBV‐CLS‐484 disrupts mitochondrial renewal and blocks TFRC‐mediated mitophagy to exert anti‐tumor activities, supporting clinical development for ALK+ ALCL.
