LRRC8A Regulates Outer Hair Cell Volume and Electromotility and is Required for Hearing

open access: yesAdvanced Science, EarlyView.
This study identifies LRRC8A‐dependent volume‐regulated anion channels (VRACs) as essential for cochlear outer hair cells' electromotility and auditory signal amplification. LRRC8A deficiency disrupts cell volume control, impairs auditory sensitivity, and causes deafness, while targeted LRRC8A re‐expression restores auditory function.

Nicotine Reprograms Aging‐Related Metabolism and Protects Against Motor Decline in Mice

open access: yesAdvanced Science, EarlyView.
Long‐term oral nicotine intake protects against age‐related motor decline in mice without eliciting systemic toxicity. Integrated multi‐organ metabolomic profiling and longitudinal gut microbiota analyses reveal that nicotine induces coordinated remodeling of glycolipid and sphingolipid metabolism, enhances NAD⁺ bioavailability, and suppresses ceramide

The DEC2‐SCN2A Axis is Essential for the Anticonvulsant Effects of Cannabidiol by Modulating Neuronal Plasticity

open access: yesAdvanced Science, EarlyView.
This study identifies DEC2 as a critical transcriptional repressor of Scn2a that directly binds E‐box motifs in its promoter to regulate neuronal plasticity and epileptogenesis. Functional manipulation of DEC2 alters seizure susceptibility in vivo. Cannabidiol enhances this repression, unveiling a novel DEC2‐SCN2A axis underlying its anticonvulsant ...

Cracking the Valence Code: Patterned Facial Kinematics and Neural Signatures of Emotional Expressions in Mice

open access: yesAdvanced Science, EarlyView.
An AI‐powered system decodes emotional valence from mouse facial expressions, identifying ear dynamics as key indicators. By integrating facial kinematics with synchronized neural recordings, this approach establishes a direct link between behavior and brain activity, advancing automated emotion recognition in animal models.

Aging and the Spectral Properties of Brain Hemodynamics

open access: yesAdvanced Science, EarlyView.
As the brain ages, its metabolic demands decline—but not uniformly. Here, spectral slope flattening of resting‐state fMRI signals is proposed as a potential biomarker of pathologic brain aging. A subset of older adults diverges from youthful spectral and metabolic patterns, with changes linked to frontal white matter pathology and regional loss of ...

Calcitonin Gene‐Related Peptide (CGRP)‐Expressing Neurons in the External Lateral Parabrachial Area Regulate Pain‐Induced Sleep Disturbances

open access: yesAdvanced Science, EarlyView.
Lynch et al., identified novel targets on the wake‐promoting pathways that can effectively treat pain‐induced sleep disturbances. The authors demonstrated that Calcitonin gene‐related peptide‐expressing neurons (CGRP) in the external lateral parabrachial nucleus (PBelCGRP) serve as the primary relay node for pain stimuli that cause sleep disruption ...
