Trichostatin A Sensitizes Tumor Cells to Apoptosis Induced by Soluble Trimeric TRAIL‐Expressing Human Mesenchymal Stromal Cells

open access: yesCancer Science, EarlyView.
Human mesenchymal stromal cells (MSCs) were engineered to secrete an isoleucine‐zippered soluble trimeric TRAIL (SCZT) and evaluated with trichostatin A (TSA) to overcome TRAIL resistance. TSA upregulated DR5 and amplified caspase‐8–dependent apoptosis in vitro; in vivo, tumor‐homing MSCs combined with TSA suppressed the growth of both TRAIL‐sensitive ...

Crucial Contribution of BACH1 to Bladder Cancer Progression via Upregulating Epithelial‐Mesenchymal Transition Pathway

open access: yesCancer Science, EarlyView.
BACH1 is associated with poor prognosis in BC patients and plays a critical role in promoting tumor progression and metastasis. Furthermore, we elucidated the mechanism by which BACH1 facilitates tumor progression and metastasis, primarily through activation of the EMT pathway. These findings suggest that BACH1 may serve as a novel prognostic biomarker

Expression of the CXCR4 S338X Variant Improves Anti‐Leukemia Efficacy of Anti‐CD19 CAR‐T Cells

open access: yesCancer Science, EarlyView.
Under normal conditions, CXCL12 stimulation induces CXCR4 degradation. In contrast, CAR19‐T cells expressing the CXCR4 S338X variant maintain receptor stability and sustain ERK and AKT signaling, which supports T‐cell effector function, migration, and survival. Therefore, the enhanced anti‐leukemic activity is conferred by CXCR4 S338X expression in CAR‐

Mechanism of ITGB2 in Osteoclast Differentiation in Osteoarthritis

open access: yesCell Proliferation, EarlyView.
In osteoclast precursors, higher ITGB1 than ITGB2 expression increases ITGA2‐ITGB1 integrin. In OA, extracellular signals upregulate ITGB2 and downregulate ITGB1, enhancing ITGA2‐ITGB2 integrin. This activates Rac1 to Rac‐GTP, driving osteoclast differentiation. Hyperactive subchondral osteoclasts accelerate OA progression.

Structural and functional characterization of the newly identified Photorhabdus laumondii tumor necrosis factor‐like lectin

open access: yesThe FEBS Journal, EarlyView.
We characterize the PLTL lectin from Photorhabdus laumondii, which adopts a unique TNF‐like fold. Binding assays reveal a preference for branched histo‐blood group oligosaccharides. Protein crystallography uncovers a novel hexameric assembly and identifies key residues involved in carbohydrate recognition.

Functional divergence of protein kinase A regulatory subunit Iβ variants: the importance of N3A motifs in PKA regulation

open access: yesThe FEBS Journal, EarlyView.
The regulatory subunit RIβ of protein kinase A occurs in two variants that differ at residue 268 (A268 versus R268). This seemingly small substitution significantly changes cAMP sensitivity, basal kinase activity, and the dynamic behavior of the regulatory domains.

Cellular Bioluminescence Imaging

Cold Spring Harbor Protocols, 2012
Bioluminescence imaging of live cells has recently been recognized as an important alternative to fluorescence imaging. Fluorescent probes are much brighter than bioluminescent probes (luciferase enzymes) and, therefore, provide much better spatial and temporal resolution and much better contrast for delineating cell structure.

Bioluminescence imaging in living organisms

Current Opinion in Biotechnology, 2005
Luciferase enzymes catalyze the emission of light from a substrate -- a phenomenon known as bioluminescence -- and have been employed as reporters of many biological functions. Luminescent reporters are much dimmer than fluorescent reporters, and therefore provide relatively modest spatial and temporal resolution. Yet, they are generally more sensitive

Bioluminescent Imaging in Bone

2008
Monitoring gene expression in vitro and in vivo, is crucial when analyzing osteogenesis and developing effective bone gene therapy protocols. Until recently, molecular analytical tools were only able to detect protein expression either in vitro or in vivo.

Bioorthogonal chemistry in bioluminescence imaging

Drug Discovery Today, 2018
Bioorthogonal chemistry has developed significant over the past few decades, to the particular benefit of molecular imaging. Bioluminescence imaging (BLI) along with other imaging modalities have significantly benefitted from this chemistry. Here, we review bioorthogonal reactions that have been used to signific antly broaden the application range of ...
