Total synthesis of brevetoxin A
Nature, 1998Brevetoxin A is the most potent neurotoxin secreted by Gymnodinium breve Davis, a marine organism often associated with harmful algal blooms known as 'red tides'. The compound, whose mechanism of action involves binding to and opening of sodium channels, is sufficiently toxic to kill fish at concentrations of nanograms per ml and, after accumulation in

Cardiovascular effects of brevetoxins in dogs
Toxicon, 1985In anesthetized spontaneously breathing dogs, brevetoxin caused dose-dependent periods of apnea and, at high doses, respiratory arrest. In artificially ventilated dogs, i.v. brevetoxin caused complex dose-related cardiovascular changes consisting of: (1) bradycardia; (2) triphasic blood pressure changes, sequentially characterized by depressor/pressor ...

Total Synthesis of Brevetoxin B
Journal of the American Chemical Society, 2005The convergent total synthesis of brevetoxin B (1) has been achieved. The intramolecular allylation of the O,S-acetal 20, prepared from the alpha-chlorosulfide 17 and the alcohol 5, was carried out using AgOTf as a Lewis acid to give the diene 21, predominantly.

Absolute configuration of brevetoxins
Journal of the Chemical Society, Chemical Communications, 1986The absolute configuration of the dinoflagellate toxins, brevetoxins, has been unequivocally determined by X-ray crystallography of a chiral dioxolane derivative.

Tetrahedron Letters, 1982
Abstract The structure of brevetoxin C (BTX-C), a polyether marine toxin from Ptychodiscus brevis , has been elucidated by comparison of its spectral data with those of brevetoxin B (BTX-B).

The chemistry of brevetoxins: A review
Toxicon, 1985The structure of the unique 'red tide' dinoflagellate neurotoxin, brevetoxin-B is presented and the experimental data supporting the chemical structure is discussed. A brief account of the other brevetoxins and their structural relationships is also presented.

Improved Synthesis of the ABCDE Fragment of Brevetoxin A
Organic Letters, 2006A second-generation synthesis of the BCDE fragment of brevetoxin A is described. Novel reactions were developed that extend the utility of the asymmetric glycolate alkylation reaction and improve scale-up to provide gram quantities of the B and E subunits. Significant improvements to the convergent assembly of the tetracycle were also realized.

Distribution of Brevetoxin to Lipoproteins in human plasma
Toxicon, 2007To better understand the distribution of brevetoxins in lipoproteins, including their role in tissue delivery and toxin elimination in humans, we examined the interaction of brevetoxin congener PbTx-3 with human lipoproteins. In a scintillation proximity assay (SPA) and microtiter equilibrium dialysis, brevetoxin bound linearly to purified human high ...

Immune effects of the neurotoxins ciguatoxins and brevetoxins
Toxicon, 2018Ciguatoxins (CTXs) and brevetoxins (PbTxs) are phycotoxins that can accumulate along the marine food chain and thus cause seafood poisoning in humans, namely "ciguatera fish poisoning" (CFP) and "neurotoxic shellfish poisoning" (NSP), respectively. CFP is characterized by early gastrointestinal symptoms and typical sensory disorders (paraesthesia, pain,

A Highly Convergent Approach to Brevetoxin A
Organic Letters, 2007Through optimized, scaled-up routes to the G and J rings of brevetoxin A, a convergent strategy based upon a Horner-Wadsworth-Emmons coupling reaction completed the GHIJ fragment. Further studies toward an alternative protecting group strategy for the GHIJ fragment elucidated a shortened synthesis.
