Attempts to Achieve Targeted Covalent Inhibition of Hsp90β
Electrophilic warheads were incorporated into Hsp90β‐selective inhibitors to covalently modify Lys58, which occupies the solvent‐exposed region of the binding pocket. Although many inhibitors display nanomolar affinity towards Hsp90β over a 24‐h period, analyses to determine covalent bond formation with this residue were inconclusive.
