The role of the BTLA-HVEM complex in the pathogenesis of autoimmune diseases

open access: yesCellular Immunology, 2022
Autoimmune diseases constitute a heterogeneous group of disorders with one common feature - the loss of immune tolerance towards autoantigens. Due to the complexity of the pathogenesis of these diseases, there are still many open questions regarding their etiology.

The BTLA-HVEM complex – The future of cancer immunotherapy

open access: yesEuropean Journal of Medicinal Chemistry
The BTLA-HVEM complex plays a pivotal role in cancer and cancer immunotherapy by regulating immune responses. Dysregulation of BTLA and HVEM expression contributes to immunosuppression and tumor progression across various cancer types. Targeting the interaction between BTLA and HVEM holds promise for enhancing anti-tumor immune responses. Disruption of

Epitope topography of agonist antibodies to the checkpoint inhibitory receptor BTLA

open access: yesStructure, 2023
B and T lymphocyte attenuator (BTLA) is an attractive target for a new class of therapeutics that attempt to rebalance the immune system by agonizing checkpoint inhibitory receptors (CIRs). Herpesvirus entry mediator (HVEM) binds BTLA in both trans- and cis-orientations.

Role of BTLA/HVEM network in development of gastric cancer

Human Immunology, 2022
The immunopathological mechanism underlying intestinal metaplasia and gastric cancer remain incompletely understood. Regarding the role of B- and T-lymphocyte attenuator (BTLA) / herpesvirus entry mediator (HVEM) in tumorigenesis, this research was conducted to determine the BTLA/HVEM expression in development of gastric cancer.

Association of a BTLA gene polymorphism with the risk of rheumatoid arthritis [PDF]

open access: yesJournal of Biomedical Science, 2006
B and T lymphocyte attenuator (BTLA) is an immuno-inhibitory receptor with the ability to deliver inhibitory signal for suppressing lymphocyte activation. To test the potential association of the human BTLA gene with the development of rheumatoid arthritis (RA), a genetic case-control association study was conducted, by using a single nucleotide ...

Balancing co-stimulation and inhibition with BTLA and HVEM

Nature Reviews Immunology, 2006
The interaction between B- and T-lymphocyte attenuator (BTLA), an inhibitory receptor whose extracellular domain belongs to the immunoglobulin superfamily, and herpesvirus-entry mediator (HVEM), a co-stimulatory tumour-necrosis factor receptor, is unique in that it is the only receptor-ligand interaction that directly bridges these two families of ...

Defining the Role of BTLA in Breast Cancer Immunosurveillance and Selective Targeting of the BTLA-HVEM-LIGHT Constimulatory System

2011
Abstract : Costimulatory (CD28, ICOS) and inhibitory (CTLA4, PD-1) molecules of the CD28 receptor family provide critical secondary signals regulating the balance between protective immunity and tissue injury. We recently cloned B- and T-lymphocyte attenuator (BTLA), the third inhibitory receptor of the CD28 family expressed on lymphocytes.

Regulation of T cell CD5 levels by BTLA

The Journal of Immunology, 2022
Abstract Many coinhibitory receptors are absent from naïve T cells and upregulated upon activation. In contrast, there are a small number of coinhibitory receptors expressed constitutively by naïve T cells, including CD5, BTLA, and VISTA. The relationship between these constitutively expressed coinhibitors is unknown.

BTLA: a new inhibitory receptor with a B7-like ligand

Trends in Immunology, 2003
The B and T lymphocyte attenuator, BTLA, is a recently discovered Ig superfamily member. BTLA engagement results in downregulation of T-cell activation, and mice deficient in BTLA show increased incidence and severity of autoimmune disorders. A novel B7 family member, B7x (B7S1, B7-H4), has been proposed as the ligand for BTLA.
