Evaluation of an accelerated Caco-2 cell permeability model
Journal of Pharmaceutical Sciences, 2000An accelerated 3-7-day Caco-2 cell permeability model was examined and compared to the traditional 21-25-day model. Caco-2 cell permeability coefficients (P(Caco-2)) of 33 structurally diverse small molecular weight compounds from apical to basolateral (AP-->BL) direction in the accelerated model were approximately twice those in the traditional model.

AbstractThe human intestinal epithelium is composed of several cell types, mainly enterocytes and goblet (mucin‐secreting) cells. This study compares the cellular response of Fe transporters in Caco‐2, HT29‐MTX, and Caco‐2/HT29‐MTX co‐culture models for Fe bioavailability. Caco‐2 cells in vitro differentiate into enterocyte‐like cells and HT29‐MTX cell

Regulation of NHE3 by nitric oxide in Caco-2 cells
American Journal of Physiology-Gastrointestinal and Liver Physiology, 2002The effect of nitric oxide (NO) on Na+/H+exchange (NHE) activity was investigated utilizing Caco-2 cells as an experimental model. Incubation of Caco-2 cells with 10−3M S-nitroso- N-acetylpenicillamine (SNAP), a conventional donor of NO, for 20 min resulted in a ∼45% dose-dependent decrease in NHE activity, as determined by assay of ...

Osmotic stress, a proinflammatory signal in Caco-2 cells
Biochimie, 2004Hyper- (450 mOsm/l) and hypoosmotic exposure (150 mOsm/l) of Caco-2 cells, a human intestinal epithelial cell line, induced a twofold- and a fivefold increase in the production of IL-8, a constitutively expressed cytokine, respectively. This was observed both in the presence or in the absence of added proinflammatory cytokines and the stimulatory ...

Stereoselectivity of the reduced folate carrier in Caco‐2 cells
Chirality, 2005AbstractStereoselectivity of the human reduced folate carrier (RFC1) was examined in Caco‐2 cells using methotrexate (l‐amethopterin or l‐MTX) and its antipode (d‐amethopterin or d‐MTX) as model substrates. The initial uptake rate of folic acid (FA) was concentration dependent, with a Km value of approximately 0.6 μM. The Eadie–Hofstee plot of the RFC1‐

Campylobacter jejuni motility and invasion of Caco-2 cells
Infection and Immunity, 1995We investigated the influence of motility on Campylobacter jejuni binding and invasion of Caco-2 cells. C. jejuni was motile in soft agar at basic (pH 8.5) and neutral pH values representative of the intestinal environment. However, C. jejuni was immobilized at pH 5.0. The inability of C.

Toxic and biochemical effects of zinc in Caco-2 cells
Journal of Inorganic Biochemistry, 2003Zinc (in relatively high concentrations) can be toxic to intestinal cells. The aim of the present study was to quanitfy cellular injury in preconfluent, colonic cancerous cells and in postconfluent, differentiating human intestinal Caco-2 cells. Cellular damage was measured by using cell proliferation, lactate dehydrogenase (LDH)-release, and apoptosis

Determination of Iron in Caco-2 cells by ET-AAS
Analytical and Bioanalytical Chemistry, 2005An electrothermal atomic absorption spectrometric method (ET-AAS) was developed for the direct determination of iron in intestinal Caco-2 cells after studying cell viability and proliferation using 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl tetrazolium bromide (MTT test).

Putrescine transport in differentiated Caco-2 cells.
Gastroenterologie clinique et biologique, 1996To evaluate the putative importance of digestive luminal polyamines, we investigated the role of oxidation in putrescine uptake and transport in differentiated Caco-2 cells grown in monolayer. Our results confirm a putrescine uptake system located both at the apical and basolateral cell membrane. Putrescine is efficiently transported and excreted in the

Rhamnolipids enhance epithelial permeability in Caco-2 monolayers
International Journal of Pharmaceutics, 2013This work aimed to evaluate the applicability of rhamnolipids as permeation enhancers for oral drugs. In this study, rhamnolipids were found to effectively increase the paracellular and transcellular transport of Transwell(®)-cultured Caco-2 cells, an in vitro model of the human small intestinal epithelium, in a concentration-dependent manner ...
