Integrin αVβ3‐targeting small‐molecule drug conjugates (SMDCs) were synthesized by conjugating a cryptophycin payload through a neutrophil elastase‐cleavable NPV‐PABC linker. RGD peptidomimetic and linker epimers served as controls for targeting and enzymatic cleavage, and the resulting conjugates displayed subnanomolar cytotoxicity in vitro.

Nitrooxylation—the installation of nitrate esters (–ONO2) has evolved from classical mixed‐acid protocols to catalytic and mechano‐, flow‐, photo‐, and electrochemical strategies. This review delineates ionic and radical manifolds for C–ONO2 bond formation, emphasizing mechanistic control, selected substrate scope, and selectivity.

Fluorogenic Sydnones for Bioorthogonal Labeling of DNA
Conjugates of sydnones with cyanine‐styryl dyes combine bioorthogonal reactivity with fluorogenicity. The reactivity with bicyclo[6.1.0]non‐4‐yne (BCN)‐modified DNA showed second‐order rate constants of up to k2 = 2,300 M−1s−1 and fluorescence turn‐on by one magnitude of order.

Control of Gemcitabine Activity With Blue and Red Light
Treating cancer with chemotherapy is generally associated with many side effects, which can be addressed by selectively releasing the desired drugs with visible light. Sixteen new blue light‐activatable cytostatics were introduced, with the gemcitabine derivative exhibiting ideal properties.

Stereochemical Control in the Matteson Reaction
The Matteson homologation is a highly versatile method for the stereoselective insertion of carbenoids into boronic esters. Through iterative application of the process, complex chiral molecules can be synthesized. The stereoselectivity of the reaction is determined by a chiral diol incorporated in the boronic ester.
