Prime Editor 7‐mediated conversion of APOE4 to APOE3 alleviates Alzheimer's disease‐associated pathology in AD mouse models and patient‐derived neurons and improves cognitive performance in vivo, supporting therapeutic genome editing as a promising strategy for APOE4‐associated neurodegeneration.

A Supramolecular Thermal Switch for Precision Pyroptosis via Host−Guest Recognition and Electrostatic Interactions

A supramolecular pyroptotic thermal switch encapsulates I3− within β‐cyclodextrin (β‐CD) and undergoes CaCO3 mineralization. Upon lysosomal acidification, CaCO3 is degraded, releasing Ca2+ and I3−. Synchronized Imaging of Hydrogen Peroxide and Hydroxyl Radical in Pyroptosis and Epilepsy

A dual‐responsive fluorescent probe, HH, enables synchronized visualization of H2O2 and •OH through spectrally resolved green and red channels. By tracking redox dynamics in pyroptosis and epilepsy models, HH reveals ROS elevation during seizure‐like pathology and supports evaluation of antioxidant and antiepileptic therapeutic interventions.

Cancer‐Associated BCL‐2 Mutants Reveal Mechanisms Towards Venetoclax Resistance

Venetoclax (VEN) resistance in chronic lymphocytic leukemia arises from diverse BCL2 mutations. We map mechanisms contributing to VEN resistance across common BCL‐2 variants. G101V and D103Y reduce drug binding and increase sequestration of pro‐apoptotic proteins. V156D blocks VEN allosterically.

NSD2 Coordinates the Neurogenic‐to‐Gliogenic Transition via H3K36me2‐Dependent Activation of the EGFR‐ERK Pathway

NSD2 coordinates the neurogenic‐to‐gliogenic transition in the developing neocortex through H3K36me2‐dependent activation of EGFR–ERK signaling. Loss of NSD2 disrupts astroglial and oligodendroglial development, whereas ERK activation rescues gliogenic defects in vitro and in vivo.
