Intermediate clinical endpoints as surrogates for overall survival after salvage prostatectomy
Objectives To evaluate biochemical recurrence‐free survival (BRFS) and metastasis‐free survival (MFS) as potential intermediate clinical endpoints (ICEs) for overall survival (OS) in patients undergoing salvage radical prostatectomy (sRP). Patients and Methods Evaluable patients were selected from a retrospective dataset, resulting in a cohort of 879 ...

Ablative radiotherapy in castration‐resistant prostate cancer
Objective To prove the oncological benefit of ablative radiotherapy in patients with up to five metastases from castration‐resistant prostate cancer (CRPC) a single‐centre randomised trial was initiated. Patients and Methods This monocentric, randomised, phase II clinical trial enrolled patients with up to five prostate‐specific membrane antigen ...

Targeting KRAS for cancer therapy
In recent years, therapeutics targeted against KRAS proto‐oncogene GTPase (KRAS)‐mutant cancers have seen significant progress. Herein we outline the biology and epidemiology of KRAS alterations at the lineage and allele levels, reviewing the clinical evidence for KRASG12C inhibition from the discovery of the recessive switch pocket to sotorasib ...

Intraindividual epigenetic heterogeneity underlying phenotypic subtypes of advanced prostate cancer
Castration-resistant prostate cancer is a heterogeneous disease with variable phenotypes commonly observed in later stages of the disease. These include cases that retain expression of luminal markers and those that lose hormone dependence and acquire ...

Abstract Cardiovascular disease (CVD) remains the leading global cause of death, driven by complex mechanisms, in which chronic inflammation plays a central role. Inflammatory pathways contribute to all stages of CVD, from endothelial dysfunction and plaque formation to erosion, rupture and myocardial injury.

ASCT2 inhibition induces ROS accumulation and activates NRF2‐dependent xCT upregulation in prostate cancer. This adaptive response partially supports residual cystine acquisition and glutathione synthesis, whereas combined ASCT2 and xCT inhibition disrupts redox compensation and predominantly promotes apoptotic cell death.

Targeting pre-existing club-like cells in prostate cancer potentiates androgen deprivation therapy
A critical knowledge gap in prostate cancer research is understanding whether castration-tolerant progenitor-like cells that reside in treatment-naïve tumors play a direct role in therapy resistance and tumor progression.

Hypericin induces autophagic AKT degradation by promoting AKT‐HSPA5 interaction in an S473 phosphorylation‐dependent manner, suppressing HCC growth. It enhances CD8+ T cell‐mediated antitumor immunity and synergizes with anti‐PD‐L1 therapy, showing favorable safety for HCC treatment. ABSTRACT Hepatocellular carcinoma (HCC) is the most prevalent primary

ABSTRACT Fathers, particularly in conservative patriarchal societies and politically complex contexts, are often underrepresented in research on child protection and out‐of‐home placements. Addressing this significant knowledge gap, this study focuses on Palestinian fathers from East Jerusalem whose young children were removed from their homes by court
