Organelle proteomics [PDF]

open access: yesCurr Protoc Protein Sci Chapter 24 (2006) Unit 24.2, 2009
This unit describes strategies for studying the proteomes of organelles, which is one example of targeted proteomics. It relies heavily on previously published units dealing with organelle preparation, protein solubilization, and proteomics techniques. A specific commentary for organelle proteomics is provided.

A long-term in vitro silicon-based microelectrode-neuron connection [PDF]

open access: yes, 1988
A novel method for long-term recording and simulation applicable to cultured neurons has been developed. Silicon-based microelectrodes have been fabricated using integrated-circuit technology and micromachining.

Virological applications of the grid-cell-culture technique

open access: yesElectron Microscopy Reviews, 1990
Whole mounts of intact virus-infected cells have been used for several decades to examine virus-cell relationships and virus structure. The general concept of studying virus structure in association with the host cell has recently been expanded to reveal interactions between viruses and the cytoskeleton. The procedure permits utilization of immuno-gold

Three-dimensional cell culture technique and pathophysiology [PDF]

open access: yesAdvanced Drug Delivery Reviews, 2014
Three-dimensional (3D) tissue constructs consisting of human cells have opened a new avenue for tissue engineering, pharmaceutical and pathophysiological applications, and have great potential to estimate the dynamic pharmacological effects of drug candidates, metastasis processes of cancer cells, and toxicity expression of nano-materials, as a 3D ...

The [2Fe‐2S] cluster of mitochondrial outer membrane protein mitoNEET has an O2‐regulated nitric oxide access tunnel

open access: yesFEBS Letters, EarlyView.
The mitochondrial outer membrane iron–sulphur ([Fe‐S]) protein mitoNEET is a target of the type‐2 diabetes drug pioglitazone. Its unknown molecular function is linked to respiratory complex activity and mitochondrial function. We discovered that O2 protects the mitoNEET [2Fe‐2S] cluster against NO oxidation and desensitization towards reduction by H2S.

Evaluation of the cell viability of human Wharton's jelly stem cells for use in cell therapy [PDF]

open access: yes, 2012
Human umbilical cord Wharton's jelly stem cells (HWJSCs) are gaining attention as a possible clinical source of mesenchymal stem cells for cell therapy and tissue engineering due to their high accessibility, expansion potential, and plasticity.

A histidine‐rich extension of the mitochondrial F0 subunit ATP6 from the ice worm Mesenchytraeus solifugus increases ATP synthase activity in bacteria

open access: yesFEBS Letters, EarlyView.
The glacier ice worm Mesenchytraeus solifugus survives year‐round at 0 °C. Its ATP6 subunit, which forms a regulatory component of the proton pore in mitochondrial ATP synthase, has a carboxy‐terminal extension not found in any other organism examined to date. Here, we show that fusion of this extension to the homologous AtpB protein in E. coli results

Refinements in primary rumen epithelial cell culture techniques [PDF]

open access: yes, 1999
The objectives of this study were to 1) determine if number of rumen epithelial cells incubated in a primary cell culture affects the rate of metabolite production; and 2) determine the optimum mode of data expression to standardize reporting criteria. A

Taurine promotes glucagon‐like peptide‐1 secretion in enteroendocrine L cells

open access: yesFEBS Letters, EarlyView.
Taurine, a sulfur‐containing amino acid, is likely taken up by enteroendocrine L cells via the taurine transporter. This process increases the levels of cytosolic ATP. The increase in intracellular Ca2+ concentrations and glucagon‐like peptide‐1 secretion through membrane depolarization is caused by the closure of ATP‐sensitive potassium channels ...

A Protocol for Achieving Adherent Cell Culture within a Microfluidic Device [PDF]

open access: yes, 2020
The goal of this study is to design a protocol for the adherent cell culture within a novel microfluidic device. Microscale cell culture protocols were developed for loading cells using poly-L-lysine to enhance adherent cell culture of murine derived NIH
