Cellular Uptake of Oligodeoxynucleotide Phosphorothioates and Their Analogs

The uptake and intracellular distribution of oligonucleotide phosphorothioates (S-oligonucleotides) and their analogs by various cells lines were studied using internally 35S-labeled oligonucleotides. Intracellular accumulation starts in the first 2 hr and reaches a maximum at about 16 hr.

SYNOPSISThe uptake of RNase by two types of amoeba and a selection of vertebrate cells has been investigated using the fluorescein‐labelled enzyme. This method is considered satisfactory for the study since the fluorescein label migrated together with the enzyme during electrophoresis, and the labelled RNase retained one half of its enzymatic activity.

AbstractPure, perfectly spherical cellulose nanoparticles with sizes of ≈80–260 nm can be prepared by dialysis starting from trimethylsilylcellulose (TMSC). The aqueous suspensions obtained are storable for several months. Subsequent covalent labeling of the cellulose nanoparticles with FITC has no influence on particle size, shape, and stability.

This chapter describes various approaches allowing the study of hyperosmolarity in the functions of 3T3-L1 adipocytes. Hyperosmolarity mimics insulin responses, such as glucose uptake and membrane ruffling, but also antagonizes these insulin effects, which can be evaluated in 3T3-L1 adipocytes.
