Nitrated Nucleotides: New Players in Signaling Pathways of Reactive Nitrogen and Oxygen Species in Plants

open access: yesFrontiers in Plant Science, 2020
Nitration of diverse biomolecules, including proteins, lipids and nucleic acid, by reactive nitrogen species represents one of the key mechanisms mediating nitric oxide (NO) biological activity across all types of organisms.

Continuous Purification of mRNA Produced by In Vitro Transcription Using High Performance Countercurrent Membrane Purification

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Messenger ribonucleic acid (mRNA) therapeutics produced by in vitro transcription must be purified to remove residual enzymes and free nucleotides. This study examines the use of high‐performance countercurrent membrane purification (HPCMP) for the purification of mRNA therapeutics based on differences in the rate of diffusion across ...

Activation of HIV-1 expression and replication by cGMP dependent protein kinase type 1-β (PKG1β)

open access: yesRetrovirology, 2007
The effect of cGMP (cyclic GMP) dependent protein kinase 1-β (PKG1-β) and cGMP analogues on transcriptional activity and replication of human immunodeficiency virus type 1 (HIV-1) was investigated.

Mechanisms associated with cGMP binding and activation of cGMP-dependent protein kinase [PDF]

open access: yesProceedings of the National Academy of Sciences, 2003
Using small-angle x-ray scattering, we have observed the cGMP-induced elongation of an active, cGMP-dependent, monomeric deletion mutant of cGMP-dependent protein kinase (Δ 1–52 PKG-Iβ). On saturation with cGMP, the radius of gyration of Δ 1–52 PKG-Iβ increases from 29.4 ± 0.1 Å to 40.1 ± 0.7

Glycoengineered Recombinant Alpha1‐Antitrypsin Results in Comparable In Vitro and In Vivo Activities to Human Plasma‐Derived Protein

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Alpha‐1‐antitrypsin (A1AT) is a multifunctional, clinically important, high‐value therapeutic glycoprotein that can be used for the treatment of many diseases, such as A1AT deficiency, diabetes, graft‐versus‐host disease, cystic fibrosis, and various viral infections. Currently, the only U.S. food and drug administration‐approved treatment for

Phosphodiesterase 5 attenuates the vasodilatory response in renovascular hypertension.

open access: yesPLoS ONE, 2013
NO/cGMP signaling plays an important role in vascular relaxation and regulation of blood pressure. The key enzyme in the cascade, the NO-stimulated cGMP-forming guanylyl cyclase exists in two enzymatically indistinguishable isoforms (NO-GC1, NO-GC2) with

Interconvertible cGMP-free and cGMP-bound forms of cGMP-dependent protein kinase in mammalian tissues.

open access: yesJournal of Biological Chemistry, 1987
Several vascular and nonvascular mammalian tissue extracts exhibited variable amounts of two peaks (peaks I and II) of cGMP-dependent protein kinase by NaCl elution of DEAE columns. When [3H]cGMP was added to the extracts before chromatography, a peak of protein-bound [3H]cGMP coeluted with peak II.

Partial Reconstitution of Photoreceptor cGMP Phosphodiesterase Characteristics in cGMP Phosphodiesterase-5 [PDF]

open access: yesJournal of Biological Chemistry, 2001
Photoreceptor cGMP phosphodiesterases (PDE6) are uniquely qualified to serve as effector enzymes in the vertebrate visual transduction cascade. In the dark-adapted photoreceptors, the activity of PDE6 is blocked via tight association with the inhibitory gamma-subunits (Pgamma).

Status and future of recombinant adeno‐associated virus vector manufacturing

open access: yesBiotechnology Progress, EarlyView.
Abstract Sixty years of adeno‐associated virus (AAV) research illustrates a trajectory marked by basic science exploration, iterative innovation, persistent challenges, a number of clinical setbacks, as well as commercial therapeutic triumphs. This continual evolution has led to recombinant AAV (rAAV) becoming a cornerstone of modern gene therapy ...
