A Mitochondria‐Targeted Nanozyme Platform for Multi‐Pathway Tumor Therapy via Ferroptosis and Cuproptosis Regulation

This work reports a mitochondria‐targeted nanozyme platform, MIL‐Cu1.8S‐TPP/FA, which induces potent cuproptosis via localized delivery of Cu1.8S nanodots to mitochondria. Surface‐anchoring these nanodots onto MIL‐88B metal‐organic frameworks enhances both passive and active tumor targeting.

Targeting Intratumoral Copper Inhibits Tumor Progression via p62‐Mediated EZH2 Degradation and Potentiates Anti‐PD‐1 Immunotherapy in Oral Squamous Cell Carcinoma

The authors find that by targeting intratumoral copper, they can enhance p62‐mediated ubiquitination of EZH2 at the Ub‐K63 site by suppressing copper binding to SMURF2, an E3 ligase of EZH2, leading to its autophagic degradation. This mechanism suppressed OSCC progression and potentiated anti‐PD‐1 immunotherapy, highlighting a potential new therapeutic

Extracellular matrix compliance plays a critical role in the regulation of integrin‐mediated adhesion and cell motility. Here, it is reported that strengthening the kindlin2‐integrin β6 interaction overcomes the dependence on extracellular substrate stiffness and promotes integrin β6‐mediated cell migration.
