Mechanisms involved in the muscle relaxing effects of STW 5 in guinea pig stomach

The phytomedicine STW 5 (Iberogast®) improves functional dyspepsia symptoms partly through proximal gastric relaxation. We found that STW 5 inhibited ORAI‐coupled store‐operated Ca2+ entry. This novel mode of action of a smooth muscle relaxing drug impact on design of drugs and approaches to treat motility disorders.

Effect of Iberogast (STW5) on tolerance to colonic gas in patients with irritable bowel syndrome: A randomized, double‐blind, placebo control clinical trial

open access: yesNeurogastroenterology &Motility, Volume 36, Issue 10, October 2024.
Using a double blind, cross‐over, placebo controlled study design, STW‐5 reduced abdominal symptoms induced by colonic gas filling in patients with IBS. The previously reported positive effects of STW‐5 in patients with IBS could be related to an improved tolerance to colonic gas contents, by reducing sensitivity to colonic loads.
