An insulin receptor mutant (Asp707 → Ala), involved in leprechaunism, is processed and transported to the cell surface but unable to bind insulin [PDF]

open access: yes, 1996
We have identified a homozygous mutation near the carboxyl terminus of the insulin receptor (IR) α subunit from a leprechaun patient, changing Asp707 into Ala. Fibroblasts from this patient had no high affinity insulin binding sites.

Enhancing CHO cell recombinant protein production using a perfusion‐directed host evolution approach

open access: yesBiotechnology Progress, EarlyView.
Abstract Clonally derived cell lines generated from Chinese hamster ovary (CHO) cells encounter numerous stressors when cultured in high‐intensity perfusion bioreactors leading to poor process performance. To circumvent this, the ability of CHO cells to adapt to different culture environments was exploited. Here host cells were selected in the presence

Isolation of Chinese hamster ovary cell mutants requiring the continuous presence of taxol for cell division

open access: yesJournal of Cell Biology, 1983
Chinese hamster ovary (CHO) cell mutants resistant to the cytotoxic effects of taxol and requiring the drug for normal growth were isolated in a single step.

A case study showing the role of hydrophobicity variants and other enriched mAb proteoforms on filterability through a virus filter with productivity improvement measures

open access: yesBiotechnology Progress, EarlyView.
Abstract A rapid assessment of manufacturability for drug candidates is crucial for advancing a prospective biotherapeutic from a candidate to a bulk drug substance. A lot‐to‐lot approach to manufacturability is adopted where each biologic batch is assessed for manufacturability as a bulk, unfractionated pool.

Ligand-induced endocytosis and nuclear localization of angiotensin II receptors expressed in CHO cells

open access: yesBrazilian Journal of Medical and Biological Research, 2001
A construct (AT1R-NF) containing a "Flag" sequence added to the N-terminus of the rat AT1 receptor was stably expressed in Chinese hamster ovary cells and quantified in the cell membrane by confocal microscopy after reaction with a fluorescein-labeled ...

Insights into centriole geometry revealed by cryotomography of doublet and triplet centrioles. [PDF]

open access: yes, 2018
Centrioles are cylindrical assemblies comprised of 9 singlet, doublet, or triplet microtubules, essential for the formation of motile and sensory cilia.

The GalNAc-type O-Glycoproteome of CHO Cells Characterized by the SimpleCell Strategy [PDF]

open access: yes, 2014
The Chinese hamster ovary cell (CHO) is the major host cell factory for recombinant production of biological therapeutics primarily because of its “human-like” glycosylation features.

Evaluation of gene editing in CHO cells using the Cas‐CLOVER system

open access: yesBiotechnology Progress, EarlyView.
Abstract Recent advances in gene editing technologies have transformed the genetic engineering of Chinese hamster ovary (CHO) hosts, enabling the development of cell lines with improved stability and productivity. In this study, we employed the programmable nuclease (PN) Cas‐CLOVER to precisely target the Glutamine synthetase (GS) locus in CHO cells. A

Improving Chinese hamster ovary host cell protein ELISA using Ella®: an automated microfluidic platform

open access: yesBioTechniques, 2020
Chinese hamster ovary (CHO) cells are a mammalian cell line used in the production of therapeutic proteins. Host cell proteins (HCPs) are process-related impurities that are derived from the host cell expression system.

Heat shock response in CHO mammalian cells is controlled by a nonlinear stochastic process. [PDF]

open access: yesPLoS Computational Biology, 2007
In many biological systems, the interactions that describe the coupling between different units in a genetic network are nonlinear and stochastic. We study the interplay between stochasticity and nonlinearity using the responses of Chinese hamster ovary (
