Chitin deacetylase, an enzyme isolated from Cryptococcus laurentii RY1, catalyzes the hydrolysis of acetamido group of N-acetyl-D-glucosamine unit of chitin. The primary objective of this study was to characterize and comprehend the activation of chitin deacetylase by DMSO. The secondary structure of the protein was determined by circular dichroism(CD).

Characterization and cloning of chitin deacetylases from Rhizopus circinans
Protein Expression and Purification, 2008Chitin deacetylase catalyzes hydrolysis of the acetamido groups of N-acetylglucosamine of chitin in fungal cell walls. Here a chitin deacetylase secreted by Rhizopus circinans was purified to homogeneity and partially characterized. The enzyme exhibits an apparent molecular weight of approximately 75kDa.

Chitin deacetylase product inhibition
Biotechnology Journal, 2010AbstractChitin deacetylase is the only known enzyme catalyzing the hydrolysis of the acetamino linkage in the N‐acetylglucosamine units of chitin and chitosan. This reaction can play an important role in enzymatic production of chitosan from chitin, or in enzymatic modification of chitosan, which has applications in medicine, pharmacy or plant ...

Chitin Deacetylases in Invertebrates
1986Chit in deacetylase activities have been characterised from fungi in those cases where the resulting chitosan forms a structural component of the wall, and from chitinoclastic bacteria. Here are two accounts of their activity in invertebrates, being apparently associated here in softening of chitinous tissues to allow their expansion.

Study on Enzymatic Characteristics of Chitin Deacetylase
Advanced Materials Research, 2011Chitin deacetylase (CDA) catalyzes the conversion of chitin to chitosan by the deacetylation of N-acetyl-D-glucosamine residues. The results of the fundamental enzymatic properties of chitin deacetylase producing from the high productivity chitin deacetylase strain Z7 show that, the optimum temperature of strain Z7 was 40°C, the optimum pH was 6.5 ...

Purification and Characterization of Chitin Deacetylase from Absidia coerulea
Journal of Biochemistry, 1995Chitin deacetylase, which releases the acetyl groups of glycol chitin was purified from a fungus, Absidia coerulea, and characterized. The enzyme was purified 516-fold to homogeneity by means of 65-80% ammonium sulfate precipitation followed by chromatography on Butyl Toyoperal-650M, Gigapite (hydroxyapatite), and DEAE Toyopearl-650M.

Detection of chitin deacetylase activity after polyacrylamide gel electrophoresis
Analytical Biochemistry, 1990Mucor racemosus and Rhizopus nigricans were used as sources of chitin deacetylases. Crude protein extracts were subjected to polyacrylamide gel electrophoresis at pH 8.9 (Davis system) or 4.3 (Reisfeld system) under native conditions. After electrophoresis, an overlay gel containing 0.1% (w/v) glycol chitin as substrate was incubated in contact with ...

Chitin deacetylase from Colletotrichum lindemuthianum
1988Publisher Summary Chitin, a β-1,4-1inked homopolymer of N-acetyl-D-glucosamine, is accompanied in the walls of various fungi by chitosan, a polymer similar to chitin by lacking the N-acetyl groups. Chitosan is most likely synthesized by partial deacetylation of the nascent chain of chitin once the latter is formed by chitin synthase from UDP-GlcNAc ...

The chitin deacetylase produced by marine strain Nitratireductor aquimarinus MCDA3-3 (NaCDA) was purified by using ammonium sulfate precipitation, Q Sepharose, and Superdex column chromatography. The purified NaCDA showed 75-fold purity, 50 U/mg specific activity with 28.5% yield. The purified CDA molecular weight was about 36 kDa.

Chitin and its derivatives have anticoagulant, antimicrobial, and antioxidant properties, but the poor solubility of chitin limits its application in different fields. In this study, site-directed mutagenesis was performed to enhance the deacetylation activity of chitin deacetylases CDA from Arthrobacter (ArCE4).
