Genetic and Functional Evidence Links Germline Biallelic Inactivating Variants in WWOX to Histological Mixed‐Type Thyroid Cancer

The role of germline WWOX loss‐of‐function variants in oncogenesis has remained poorly characterized. This study provides the first comprehensive genetic and functional evidence that such variants promote tumorigenesis through the disruption of multiple tumor‐suppressive mechanisms.

SV2B Promotes the Progression of TFE3‐Rearranged Renal Cell Carcinoma by Interacting with HERC2 to Impede the Degradation of NF‐κB Subunits

TFE3‐rearranged renal cell carcinoma (TFE3‐RCC) lacks clearly defined diagnostic and therapeutic targets. SV2B, a TFE3 target gene, promotes TFE3‐RCC progression by activating the NF‐κB pathway. Targeted inhibition with padsevonil or Withaferin A effectively suppresses tumor progression.

TMEM131‐Mediated Soluble TRAIL Triggered Type II Alveolar Epithelial Cell Senescence in Radiation‐Induced Lung Injury

TMEM131 recruits the COPII complex to accelerate TRAIL transportation from endoplasmic reticulum to Golgi apparatus, and promotes soluble TRAIL secretion. TRAIL inhibits mitophagy and induces senescence through DR5 receptor in type II alveolar epithelial cells, ultimately driving radiation‐induced lung injury (RILI) progression.

Punicalagin Enhances Autophagy Through Sirtuin 1/FoxO3a Axis to Inhibit Intracellular Mycobacterium Abscessus Infection

Punicalagin, a pomegranate molecule, helps the body fight Mycobacterium abscessus. It preferentially boosts interstitial macrophages in the lung, stabilizing mitochondria and switching on the autophagy via SIRT1/FoxO3a, which lowers bacterial load without directly killing bacteria.

ALDOA‐depletion‐mediated glycolytic reprogramming promotes autophagy‐mediated degradation of MBOAT2, increasing the ratio of phospholipids containing pro‐ferroptotic polyunsaturated fatty acids over anti‐ferroptotic monounsaturated fatty acids, and thus sensitizing cancer cells to ferroptosis.
