Mechanistic studies of an antibody with chorismate mutase activity

Abstract Antibody 1F7 catalyzes the rearrangement of chorismate to prephenate. Its kinetic parameters are unaffected by changes in pH and display no solvent isotope effect or effects from addition of various cationic salts. These results are consistent with high-resolution structural information of 1F7 bound to a transition state analog.

Abstract The synthesis of seven bi- and tricyclic chorismate analogues 9 - 15 and their biological evaluation as mutase inhibitors is described. A tandem Cope rearrangement/Diels-Alder cycloaddition strategy was employed to prepare tricyclo[3.3.1.02,7]non-3-enes functionalized with carboxylic and/or phosphonic acid groups.
