METTL10‐Mediated PIAS3 Methylation Links Purine Metabolism to Gastric Cancer Progression

open access: yesAdvanced Science, EarlyView.
METTL10‐mediated methylation of PIAS3 at K442 inhibits its SUMO E3 ligase activity toward MITF. This reduces MITF SUMOylation and subsequent RNF4‐mediated ubiquitination, leading to MITF protein accumulation and gastric cancer progression. Abstract Metabolic dysregulation plays a significant role in the development of gastric cancer (GC).

Variations in flanking or less conserved positions of Reb1 and Abf1 consensus binding sites lead to major changes in their ability to modulate nucleosome sliding activity

open access: yesBiological Research
Background Maintenance of nucleosome-free regions at gene regulatory regions conform a relevant aspect within chromatin dynamics. In the yeast Saccharomyces cerevisiae, Reb1 and Abf1 are among the transcriptions factors that perform this molecular ...

Epstein‐Barr Virus Expressed Long Non‐Coding RNA (lncBARTs) Regulate EBV Latent Genome Replication

open access: yesAdvanced Science, EarlyView.
EBV produces abundant level of lncBARTs, which are essential for maintaining viral genome replication in EBV‐associated cancers. LncBARTs interact with a complex comprising BRD4, CTCF and viral protein EBNA1 at EBV oriP region. This interaction tethers oriP to host chromosomes, facilitating EBV episome replication.

PM2.5 Induce Endothelial‐Mesenchymal Transition and Cardiac Fibrosis via the NCOA4‐Mediated Ferritinophagy

open access: yesAdvanced Science, EarlyView.
This study primarily reveals that PM2.5‐derived Fe‐containing particles, particularly magnetite, are specifically enriched in the heart. Here, they interact with NCOA4 in endothelial cells, disrupt iron homeostasis by enhancing ferritinophagy, and subsequently trigger the process of EndMT through the NCOA4/KLF5 pathway.

BHLHE40 Cooperates with GATA2/3 to Control Human Syncytiotrophoblast Lineage Differentiation

open access: yesAdvanced Science, EarlyView.
BHLHE40 interacts directly with GATA2 and GATA3 and co‐occupies genomic loci critical for STB differentiation and function. BHLHE40 also affects GATA2/3 binding at enhancer and promoter regions. Moreover, BHLHE40 is potentially involved in genome‐wide remodeling of H3K4me3 and H3K27ac chromatin landscapes through GATA2/3 recruitment, activating ...

Tumor Intrinsic METTL5 Modulates ATF4 Translation to Prevent T Cell‐Induced Ferroptosis in Ovarian Cancer

open access: yesAdvanced Science, EarlyView.
An integrative analysis combining in vitro genome‐wide immune screens, in vivo ICB screens, and clinical data mining identifies METTL5 as a key tumor‐intrinsic immune factor. Mechanistic studies further reveal the importance of the METTL5‐ATF4‐SLC7A11/SLC3A2‐ferroptosis axis in modulating antitumor immune responses. These findings highlight METTL5 as a
