Targeting Casein Kinase 1 (CK1) in Hematological Cancers [PDF]

open access: yesInternational Journal of Molecular Sciences, 2020
The casein kinase 1 enzymes (CK1) form a family of serine/threonine kinases with seven CK1 isoforms identified in humans. The most important substrates of CK1 kinases are proteins that act in the regulatory nodes essential for tumorigenesis of hematological malignancies. Among those, the most important are the functions of CK1s in the regulation of Wnt

Newly Developed CK1-Specific Inhibitors Show Specifically Stronger Effects on CK1 Mutants and Colon Cancer Cell Lines [PDF]

open access: yesInternational Journal of Molecular Sciences, 2019
Protein kinases of the CK1 family can be involved in numerous physiological and pathophysiological processes. Dysregulated expression and/or activity as well as mutation of CK1 isoforms have previously been linked to tumorigenesis. Among all neoplastic diseases, colon and rectal cancer (CRC) represent the fourth leading cause of cancer related deaths ...

Purification of CK1 by affinity chromatography on immobilised axin

open access: yesProtein Expression and Purification, 2007
Members of the Casein Kinase 1 (CK1) family are implicated in the regulation of a variety of physiological processes like development and circadian rhythm, as well as in diseases like cancer and Alzheimer's disease. From that perspective, CK1 family members are interesting targets for potential chemotherapy.

Isoform specific phosphorylation of p53 by protein kinase CK1

open access: yesCellular and Molecular Life Sciences, 2009
The ability of three isoforms of protein kinase CK1 (alpha, gamma(1), and delta) to phosphorylate the N-terminal region of p53 has been assessed using either recombinant p53 or a synthetic peptide reproducing its 1-28 sequence. Both substrates are readily phosphoylated by CK1delta and CK1alpha, but not by the gamma isoform.

Kinase domain autophosphorylation rewires the activity and substrate specificity of CK1 enzymes

Molecular Cell, 2022
CK1s are acidophilic serine/threonine kinases with multiple critical cellular functions; their misregulation contributes to cancer, neurodegenerative diseases, and sleep phase disorders. Here, we describe an evolutionarily conserved mechanism of CK1 activity: autophosphorylation of a threonine (T220 in human CK1δ) located at the N ...

Protein Kinase CK1 from Trypanosoma cruzi

Journal of Protein Chemistry, 2003
A protein kinase activity, which uses casein as a substrate, has been purified to homogeneity from the epimastigote stage of Trypanosoma cruzi, by sequential chromatography on Q sepharose, heparin sepharose, phenyl sepharose, and alpha-casein agarose.

Differential Phosphorylation of Occludin and Tricellulin by CK2 and CK1

Annals of the New York Academy of Sciences, 2009
In epithelial and endothelial cell layers tight junctions form selective apicolateral paracellular barriers separating luminal and extracellular spaces from the underlying tissues. Within the tight junctions the tetraspan transmembrane proteins occludin, claudins, and tricellulin form anastomosing strands of protein complexes, which interconnect ...
