Clathrin coat controls synaptic vesicle acidification by blocking vacuolar ATPase activity

open access: yeseLife, 2018
Newly-formed synaptic vesicles (SVs) are rapidly acidified by vacuolar adenosine triphosphatases (vATPases), generating a proton electrochemical gradient that drives neurotransmitter loading.

Human erythrocyte clathrin and clathrin-uncoating protein.

open access: yesJournal of Biological Chemistry, 1985
Clathrin, a Mr = 72,000 clathrin-associated protein, and myosin were purified in milligram quantities from the same erythrocyte hemolysate fraction. Erythrocyte clathrin closely resembled brain clathrin in several respects: (a) both are triskelions as visualized by electron microscopy with arms 40 nm in length with globular ends and a flexible hinge ...

A 3D Biofabricated Disease Model Mimicking the Brain Extracellular Matrix Suitable to Characterize Intrinsic Neuronal Network Alterations in the Presence of a Breast Tumor Disseminated to the Brain

open access: yesAdvanced Functional Materials, EarlyView.
A 3D disease model is developed using customized hyaluronic‐acid‐based hydrogels supplemented with extracellular matrix (ECM) proteins resembling brain ECM properties. Neurons, astrocytes, and tumor cells are used to mimic the native brain surrounding.

Clathrin-mediated endocytosis is a candidate entry sorting mechanism for Bombyx mori cypovirus

open access: yesScientific Reports, 2018
Bombyx mori cypovirus (BmCPV), a member of the Reoviridae, specifically infects silkworms and causes extensive economic losses to the sericulture industry. To date, the entry mechanism of BmCPV into cells is unclear.

Conformation Switching of Clathrin Light Chain Regulates Clathrin Lattice Assembly [PDF]

open access: yesDevelopmental Cell, 2010
Clathrin-coated vesicle formation is responsible for membrane traffic to and from the endocytic pathway during receptor-mediated endocytosis and organelle biogenesis, influencing how cells relate to their environment. Generating these vesicles involves self-assembly of clathrin molecules into a latticed coat on membranes that recruits receptors and ...

Engineering a Sonotherapeutic RBC Membrane‐Derived Nanoparticle Platform for the Treatment of Liver Cancer

open access: yesAdvanced Functional Materials, EarlyView.
Herein, an RBC membrane‐derived nanoparticle (CMN‐ICG) is engineered to efficiently deliver a sonosensitizing agent, indocyanine green (ICG), for sonotherapy of hepatocellular carcinoma (HCC). CMN‐ICG exhibits excellent cytocompatibility, significantly enhances hepatocyte uptake, and produces excessive reactive oxygen species (ROS) upon ultrasound ...

From RNA to DNA: How Cargo Identity Reprograms Lipid Nanoparticle Architecture and Function

open access: yesAdvanced Healthcare Materials, EarlyView.
The evolution of lipid nanoparticles (LNPs) spans from RNA‐LNPs, used in mRNA vaccines, to DNA‐LNPs, ideal for gene therapies. Emerging bionano architectures, decorated with DNA and plasma proteins, pave the way for advanced DNA‐based therapies that are more stable, targeted, and customizable.

Intrinsically Disordered Protein‐Inspired Nanovector‐Based Coacervates for the Direct Cytosolic Transport of Biomacromolecules

open access: yesAdvanced Materials, EarlyView.
Intrinsically disordered protein‐inspired nanovectors (IDP‐NVs) form stable nanocoacervates (NCs) with diverse biomacromolecules. In situ conformational changes confer stability and adaptability to NCs under dynamically changing physiological conditions.

Potent Liver‐Tropic mRNA Lipid Nanoparticles: ApoE‐Mediated Delivery Through a Low‐Density Lipoprotein Receptor Independent Uptake Mechanism

open access: yesAdvanced Materials, EarlyView.
Helper and ionizable lipids play a crucial role in determining ApoE binding and subsequent liver tropism and LDLR‐mediated uptake. Ionizable lipids primarily govern the LDLR‐independent uptake pathway. This complementary interplay between lipid components ultimately governs LNP delivery performance and therapeutic efficacy in the liver.

Cargo regulates clathrin-coated pit invagination via clathrin light chain phosphorylation [PDF]

open access: yesJournal of Cell Biology, 2018
Clathrin light chains (CLCs) control selective uptake of a range of G protein–coupled receptors (GPCRs), although the mechanism by which this occurs has remained elusive thus far. In particular, site-specific phosphorylation of CLCb controls the uptake of the purinergic GPCR P2Y12, but it is dispensable for the constitutive uptake of the transferrin ...
