Abstract The methods and results of inoculating callus cultures of rye( Secale cereale ) with ergot ( Claviceps purpurea ) are described. The fungus grows intercellularly and invades some cells, but does not produce alkaloids.

Summary The biosynthetic pathway of ergotoxine alkaloids (ergocornine, ergokryptine) was studied with Claviceps purpurea strain Pepty 695. Lysergic acid-U- 14 C is specifically incorporated into the lysergic acid moiety of ergotoxins. Lysergic acid amide is no intermediate in the formation of peptide type ergot alkaloids.

The development of infection of the ovary of Newal barley (Hordeum vulgare L.) by the ergot fungus, Claviceps purpurea (Fr.) Tul., was traced from the time of inoculation until formation of the sclerotia. Penetration took place within 24 hours near the base of the ovule.

Sclerotium size and germination in Claviceps purpurea

The relationship between the size of the Sclerotium and its behaviour during germination was investigated. Large sclerotia showed higher percentage germination levels at any given time during germination and gave rise to taller clavae than did smaller ones.

Proteinases of various Claviceps purpurea strains.

Extra- and intracellular proteinases from various Claviceps purpurea strains grown in submerged culture have been studied. A maximum level of intracellular proteinases was observed on the 6th day of culture growth, whereas extracellular activity continued to increase throughout the culture growth.
