Application of oxygen vectors to Claviceps purpurea cultivation
Applied Microbiology and Biotechnology, 2001The application of a two-phase fermentation system for the production of ergot peptide alkaloids by Claviceps purpurea is described. Perfluorocarbons (PFC) are used as oxygen vectors in Claviceps fermentation for the first time. In shake-flask cultivations, the inclusion of PFC in the medium brings about a five-fold increase in the total alkaloid ...

Semicontinuous cultivation of immobilized Claviceps purpurea
Applied Microbiology and Biotechnology, 1984Mycelia of Claviceps purpurea CBS 164.59 were immobilized in 2%, 4%, and 8% calcium alginate. Alkaloid production by free cells declined after 60 days, while immobilized cells retained their activity for 200 days. The cumulative alkaloid production for all fermentation cycles using 8% calcium alginate immobilized mycelia was 25 times higher than that ...

Extrachromosomal genetics of Claviceps purpurea
Current Genetics, 1983In order to establish a system for gene cloning in the ergot fungus Claviceps purpurea mitochondrial DNA was purified from two wild strains and four descendants of production strains. A characterization of the mt DNA with respect to buoyant density, molecular weight (45 kb corresponding to acontour lenght of 14 μm) showed no differences.

INFECTION OF BARLEY BY CLAVICEPS PURPUREA
Canadian Journal of Botany, 1958The development of infection of the ovary of Newal barley (Hordeum vulgare L.) by the ergot fungus, Claviceps purpurea (Fr.) Tul., was traced from the time of inoculation until formation of the sclerotia. Penetration took place within 24 hours near the base of the ovule.

REACTION OF CULTIVATED CEREALS TO CLAVICEPS PURPUREA
Canadian Journal of Plant Science, 1976The influence of inoculum concentration on the reaction induced in species and cultivars of commercial cereals by two isolates of ergot (Claviceps purpurea (Fr.) Tul.) was investigated. Differences were observed among and within species with respect to each of the three components of the disease reaction, i.e., frequency of sclerotia, size of ...

Development of stromata in Claviceps purpurea
Transactions of the British Mycological Society, 1968Observations on the development of Claviceps purpurea (Fr.) Tul. stromata showed that extension growth occurred only at the stalk apex. The rate of stalk elongation was greater in darkness than in diurnal conditions or continuous light and positive phototropic alignment of the clavae involved enhanced extension growth of hyphae on the shaded side of ...

Sclerotium size and germination in Claviceps purpurea
Transactions of the British Mycological Society, 1966The relationship between the size of the Sclerotium and its behaviour during germination was investigated. Large sclerotia showed higher percentage germination levels at any given time during germination and gave rise to taller clavae than did smaller ones.

Transformation of Claviceps purpurea using a bleomycin resistance gene
Applied Microbiology and Biotechnology, 1989To develop a DNA-mediated transformation system for Claviceps purpurea a vector was constructed using a bleomycin-resistance gene (bleoR) fused in frame to the Aspergillus nidulans trp C promoter as a dominant selection marker. The construct was shown to be functional in Aspergillus nidulans and Aspergillus niger and used to transform a wild strain of ...

Polygalacturonase is a pathogenicity factor in the Claviceps purpurea/rye interaction
Fungal Genetics and Biology, 2002Claviceps purpurea is a biotrophic, organ-specific pathogen of grasses and cereals, attacking exclusively young ovaries. We have previously shown that its mainly intercellular growth is accompanied by degradation of pectin, and that two endopolygalacturonase genes (cppg1/cppg2) are expressed throughout all stages of infection.

Transcripts and translation products of a mitochondrial plasmid of Claviceps purpurea
Current Genetics, 1992Expression of the linear mitochondrial (mt) plasmid pClK1 of strain K of the ascomycete Claviceps purpurea was studied at the RNA and protein level. Using strand-specific probes two major transcripts were detected, corresponding to ORF1 and ORF2 of the plasmid, which most likely code for a DNA-polymerase and an RNA-polymerase, respectively.
