Prophage Activation Triggers Extracellular Synthesis of DNA in Bacterial Biofilms via Rolling Circle Replication

open access: yesAngewandte Chemie, EarlyView.
Shewanella oneidensis and Bacillus subtilis, both harboring prophages, reveal a novel biosynthetic route to scalable and programmable DNA superstructures in flower‐ and wire‐like shapes, respectively. Activation of prophages is achieved via 3‐hour starvation and monitored using reporter strains.

Isolated Hepatocytes versus Hepatocyte Spheroids: In Vitro Culture of Rat Hepatocytes

open access: yesCell Transplantation, 2005
The use of hepatocytes that express liver-specific functions to develop an artificial liver is promising. Unfortunately, the loss of specialized liver functions (dedifferentiation) is still a major problem.

Intrasplenic Thymus Organogenesis from Injectable Tissue Fragments Restores Functional T‐Cell Immunity

open access: yesAdvanced Science, EarlyView.
Clinical intramuscular thymus transplantation yields only short‐lived efficacy and marginal therapeutic benefits. Benefiting from the spleen's intrinsic strengths—rapid vascular perfusion, abundant developmental factors, and resident progenitors—the intrasplenic thymic grafts achieve robust thymic regeneration and substantial T‐cell reconstitution ...

Macrophage PABPC4‐SPP1 Axis Orchestrates Immunosuppression in Colorectal Cancer

open access: yesAdvanced Science, EarlyView.
In the CRC microenvironment, macrophage PABPC4 binds to the 3′UTR of SPP1 mRNA to stabilize its expression, thereby sustaining M2‐like immunosuppressive macrophage polarization. Concurrently, this axis suppresses CD8+ T cell effector functions via CD44 signaling, collectively fostering an immunosuppressive niche that drives tumor progression.

Synovium‐On‐A‐Chip: Simulating the Microenvironment of the Rheumatoid Arthritis Synovium via Multicell Interactions to Target Fibroblast‐Like Synoviocytes

open access: yesAdvanced Science
Fibroblast‐like synoviocytes (FLSs) play pivotal roles in the synovial inflammation of rheumatoid arthritis (RA) and are promising therapeutic targets. Currently, few in vitro models effectively mimic the characteristics of RA FLSs.

DSOF: A Rapid Method to Determine the Abundance of Microalgae and Methanotrophic Bacteria in Coculture Using a Combination of Differential Sedimentation, Optical Density, and Fluorescence

open access: yesBioengineering
Cocultivation of microalgae and aerobic methanotrophs represents an emerging biotechnology platform to produce high-protein biomass, yet quantifying individual species in mixed cultures remains challenging.

Chemotherapy‐Activated GSK3β‐DNMT1 Signaling Upregulates CD47 to Evade Macrophage Phagocytosis and Drive Temozolomide Resistance in Glioblastoma

open access: yesAdvanced Science, EarlyView.
Temozolomide treatment activates GSK3β, driving DNMT1 phosphorylation, destabilization, and CD47 promoter hypomethylation in glioblastoma. This epigenetic shift upregulates CD47, enabling TMZ‐treated GBM cells to evade macrophage phagocytosis, survive chemotherapy, and acquire resistance.

Development of a direct contact astrocyte-human cerebral microvessel endothelial cells blood–brain barrier coculture model

open access: yes, 2017
ObjectivesIn conventional in-vitro blood–brain barrier (BBB) models, primary and immortalized brain microvessel endothelial cell (BMEC) lines are often cultured in a monolayer or indirect coculture or triculture configurations with astrocytes or ...

Nanofabricated Biomaterials Model Tumor Extracellular Matrix Topography

open access: yesAdvanced NanoBiomed Research, EarlyView.
The unique structure and composition of the tumor microenvironment and its associated extracellular matrix (ECM) exert profound effects on cancer cell behavior that are difficult to model in vitro. This perspective discusses the utility of nanofabricated biosubstrates, such as the novel collagen‐coated nanogrooved array, for recapitulating the ...
