SP3 and DEP1 Orchestrate Panicle Architecture by Jointly Regulating APO2 Expression in Rice

open access: yesAdvanced Science, EarlyView.
The transcription factor SP3 enhances inflorescence meristem activity and interacts with the cysteine‐rich domain of DEP1. This interaction alters DEP1's subcellular localization between the plasma membrane and nucleus, and suppresses SP3‐mediated transcriptional activation of APO2.

SCORCH2: A Generalized Heterogeneous Consensus Model for High‐Enrichment Interaction‐Based Virtual Screening

open access: yesAdvanced Science, EarlyView.
Researchers developed SCORCH2, an advanced machine‐learning scoring function for virtual screening that combines two complementary XGBoost models to re‐evaluate protein‐ligand binding plausibility. The method demonstrates superior performance on standard benchmarks, shows excellent generalization to previously unseen protein targets, and provides ...

Real‐Time Detection of Reduced Nitroreductase with a Reversible Fluorescent Probe

open access: yesAdvanced Science, EarlyView.
The first reversible binding probe (rNTRp) is presented that selectively detects the nitroreductase enzyme's active, reduced form (red‐NTR) with a fluorescence turn‐on response. Contrary to the conventional reaction‐based probes known to date, which provide only static information on the accumulated enzyme activity at a given time, rNTRp allows ...

Enhancing the β‐Oxidation‐Like Pathway for the Optimal Production of the Immunosuppressant Mycophenolic Acid

open access: yesAdvanced Science, EarlyView.
The most elaborate known example of β‐oxidation‐like pathway recruitment in fungi is reported, involving a 10‐step oxidative chain‐shortening cascade catalyzed by five dedicated enzymes to generate the pharmacophore of mycophenolic acid (MPA)—a novel clinical immunosuppressant. Our findings illustrate how nature repurposes catabolic enzymes for complex
