Complement Components C4A and C4B in Human Lupus

2005
For over half a century low serum complement C4 concentrations have been recognized as a manifestation of systemic lupus erythematosus (SLE). However, the role of C4 in human SLE remains elusive. This is partly because of the unusually complex C4 genetics with frequent variations in gene number, gene size, protein isoforms, and expression levels.

Human Complement Components C4A and C4B Genetic Diversities: Complex Genotypes and Phenotypes

Current Protocols in Immunology, 2005
AbstractThis unit describes methods that can accurately determine the genotypes and phenotypes of human complement components C4A and C4B. Specifically, they allow investigators to determine how many C4 genes are present in a diploid genome of a human subject and to quantify how many of them encode C4A proteins and how many of them encode C4B proteins.

DR3 and nonDR3 associated complement component C4A deficiency in systemic lupus erythematosus

Clinical Immunology and Immunopathology, 1991
The molecular basis of complement component C4A deficiency in white U.S. and Mexican patients with systemic lupus erythematosus (SLE) was studied. Genomic DNA from SLE patients and non-SLE controls was analyzed for restriction fragments using HindIII and a 5' C4 cDNA probe.

Allelic distribution of complement components BF, C4A, C4B, and C3 in Psoriasis vulgaris

Immunology Letters, 2002
Psoriasis vulgaris is a multifactorial disease; the strongest association was established with the HLA complex. The actual disease-predisposing gene(s) has not been identified yet, but several genes from this region were examined in addition to HLA-C and -B.

Complement C3a and C4a Increased in Plasma of Patients with Aspirin-induced Asthma

American Journal of Respiratory and Critical Care Medicine, 2006
Aspirin-induced asthma (AIA) is a distinct clinical syndrome that affects up to 10% of adults with asthma. Although eicosanoid metabolites appear to play an important role in AIA, the exact pathogenic mechanism for the syndrome remains obscure. In addition, the proposed mechanism fails to explain why aspirin does not cause bronchoconstriction in all ...

Complexes of IgG molecules and C3a and C4a complement components in human serum

European Journal of Immunology, 1992
AbstractA heterogeneous group of proteins was separated from human serum IgG using 5 M guanidine hydrochloride solution in 0.1 M acetic acid. The protein mixture was fractionated by reverse‐phase high‐performance liquid chromatography and two proteins were isolated and identified as the C3a and C4a complement components (anaphylatoxins) according to ...

Complement Factors C3a, C4a, and C5a in Chronic Obstructive Pulmonary Disease and Asthma

American Journal of Respiratory Cell and Molecular Biology, 2004
Studies of animal models have shown that the activation of the complement system could have a role in chronic obstructive pulmonary disease (COPD) and asthma by promoting inflammation and enhancing airway hyperresponsiveness. We sought to determine whether the levels of complement factors C3a, C4a, and C5a are elevated at the site of inflammation in ...

Comparative Structural Anatomy of the Complement Anaphylatoxin Proteins C3a, C4a and C5a

Enzyme, 1986
The anaphylatoxins are a family of proteins produced during the course of complement activation as the result of cleavage by specific serine proteases. These proteins are involved in a variety of biological functions, including inflammation. Comparative modeling techniques have been used to produce structures for C4a and C5a from the crystal structure ...

Rapid quantification of human complement component C4A and C4B genes by capillary gel electrophoresis

ELECTROPHORESIS, 2006
AbstractComplement component 4 (C4) is an important plasma protein playing a major role in the human defense mechanism against infectious diseases and inflammatory processes. The C4A and C4B genes, encoding the two isoforms of complement 4, are located in the nuclear serine/threonine protein kinase‐C4A or B gene‐cytochrome 21‐hydroxylase‐tenascin X ...

Activated complement components C3a and C4a in cerebrospinal fluid and plasma following subarachnoid hemorrhage

Journal of Neurosurgery, 1989
✓ The cerebrospinal fluid (CSF) and plasma levels of the complement components C3a and C4a in 40 patients suffering from subarachnoid hemorrhage (SAH) were quantitated by radioimmunoassay. Serial measurements of the lumbar CSF levels revealed that the C3a and C4a levels were significantly elevated in the initial stage of SAH, but decreased rapidly ...
