While X‐ray crystallography has been the primary method for determining the structures of metalloproteins, cryoEM has taken over this role, illustrated by [4Fe4S]‐cluster containing proteins deposited in the Protein Data Bank. What will be the role for X‐ray crystallography as cryoEM and machine learning methods dominate the initial structural analysis?

Abstract BACKGROUND Adeno‐Associated Virus vectors are widely used in gene therapy due to their safety and efficacy. Despite the interest in their therapeutical application, AAV purification remains a significant challenge. Density gradient ultracentrifugation using cesium chloride, sucrose, or iodixanol gradients are common methods for AAV ...

Linear polyglycerol is proposed as a PEG‐alternative in mRNA‐loaded lipid nanoparticles to limit activation of anti‐PEG antibodies and reduce immunogenicity. Resulting nanoparticles may provide similar gene delivery capabilities, while preventing binding of anti‐PEG antibodies compared to PEGylated counterparts.

Mass Spectrometry Structural Proteomics Enabled by Limited Proteolysis and Cross‐Linking
ABSTRACT The exploration of protein structure and function stands at the forefront of life science and represents an ever‐expanding focus in the development of proteomics. As mass spectrometry (MS) offers readout of protein conformational changes at both the protein and peptide levels, MS‐based structural proteomics is making significant strides in the

Recent advances in noncanonical inhibition mechanisms of anti‐CRISPR proteins
Abstract The CRISPR‐Cas system constitutes an adaptive immune mechanism in prokaryotes that defends against mobile genetic elements. Within the perpetual co‐evolutionary arms race between bacteria and their viral predators, bacteriophages encode anti‐CRISPR (Acr) proteins that use sophisticated molecular strategies to sabotage CRISPR‐Cas function ...
