Low reproducibility and inefficient gene transfer hamper clinical manufacturing of chimeric antigen receptor regulatory T cells (CAR Tregs). Synthetic gene circuits regulated by bezafibrate are developed to express tumor antigens and PD‐L1 nanobody (PD‐L1nb), enhancing cancer immunotherapy. Delivered via HEK293T cells, ESCs, or directly injected the plasmids containing the circuit into the tumor, the circuits reduce tumor growth, boost CD8+ T cells, and minimize T cell exhaustion.
