Nanoneedle‐Based Electroporation for Efficient Manufacturing of Human Primary Chimeric Antigen Receptor Regulatory T‐Cells

open access: yesAdvanced Science, EarlyView.
Low reproducibility and inefficient gene transfer hamper clinical manufacturing of chimeric antigen receptor regulatory T cells (CAR Tregs). Nanoneedle mediated electroporation generates primary human CAR Tregs under good manufacturing practice conditions with superior efficiency compared to viral transduction.

Host Plasma Microenvironment in Immunometabolically Impaired HIV Infection Leads to Dysregulated Monocyte Function and Synaptic Transmission Ex Vivo

open access: yesAdvanced Science, EarlyView.
Network‐based risk stratification of people living with HIV (PWH) on prolonged therapy using multi‐omics data identifies 44% of the patients are at risk of immunometabolic complications. A senescence‐associated myeloid cell‐driven plasma microenvironment drives immunometabolic dysregulation in at‐risk PWH.

Targeted STING Activation Using Modified Ultrasound‐Responsive Microbubbles Enhances Immune Checkpoint Blockade Against Melanoma

open access: yesAdvanced Science, EarlyView.
This study explores the use of ultrasound contrast agents loaded with Stimulator of Interferon Genes (STING) agonists as an approach to activate the body's immune system against cancer. Termed Microbubble‐assisted UltraSound‐guided Immunotherapy of Cancer (MUSIC), this strategy effectively inhibits tumor growth and improves survival in mouse models of ...

Circulating and Tumor-Infiltrating Foxp3+ Regulatory T Cell Subset in Chinese Patients with Extranodal NK/T Cell Lymphoma

open access: yesInternational Journal of Biological Sciences, 2011
Foxp3+ regulatory T lymphocytes (Tregs) usually act as an immune suppressor and correlate with poorer survival in malignancies. This study aims to investigate the distribution and characterization of Foxp3+ subset in peripheral blood mononuclear cells ...

Engineered Genetic Circuits Activated by Bezafibrate Improve ESC‐Based TAA Cancer Vaccine Efficacy and PD‐L1 Nanobody Therapy

open access: yesAdvanced Science, EarlyView.
Synthetic gene circuits regulated by bezafibrate are developed to express tumor antigens and PD‐L1 nanobody (PD‐L1nb), enhancing cancer immunotherapy. Delivered via HEK293T cells, ESCs, or directly injected the plasmids containing the circuit into the tumor, the circuits reduce tumor growth, boost CD8+ T cells, and minimize T cell exhaustion.

A necroptosis‐related gene signature for predicting prognosis, immune landscape, and drug sensitivity in hepatocellular carcinoma

open access: yesCancer Medicine, Volume 11, Issue 24, Page 5079-5096, December 2022., 2022
Necroptosis is a newly discovered form of cell necrosis that plays a vital role in cancer development. We identified a necroptosis‐related gene signature based on the TCGA cohort, which was validated externally in the ICGC‐LIRI‐JP and GSE14520 cohorts.

Semiconducting Open‐Shell Radicals for Precise Tumor Activatable Phototheranostics

open access: yesAdvanced Science, EarlyView.
A versatile phototheranostics nanoplatform of tumor‐specific activatable semiconducting open‐shell radicals for precise chemodynamic/photodynamic therapy (CDT/PDT) guided by “Turn On” second near‐infrared (NIR‐II) window fluorescence imaging (FLI) and PA imaging (PAI) through molecular engineering strategies. Abstract Semiconducting open‐shell radicals

Tumor‐associated macrophages regulate the function of cytotoxic T lymphocyte through PD‐1/PD‐L1 pathway in multiple myeloma

open access: yesCancer Medicine, Volume 11, Issue 24, Page 4838-4848, December 2022., 2022
Tumor‐associated macrophages(TAMs) are a vital constituent of the tumor microenvironment and involved in the immunological escape of vicious clonal plasma cells of myeloma patients. Our research shows that CSF1R can regulates the production of TAMs while the PD‐1/PD‐L1 pathway regulates the effect of TAMs on cytotoxic T lymphocyte. It may provide a new
