Engineered mesoporous silica nanoparticles (Silicasomes) enable systemic delivery of the STING agonist ADU‐S100, overcoming the instability and toxicity that limit cyclic dinucleotides to local administration. By enhancing tumor accumulation, activating systemic antitumor immunity, and remodeling the tumor immune microenvironment, these nanoparticles ...

Tumor metastasis results from complex interactions between cancer cells and mechanical microenvironments. We propose a “nucleus‐centered, cross‐stage mechanical signal decoding” model, highlighting how nuclear mechanosensors interpret forces at different stages.

Cyclic di-GMP (c-di-GMP) is a second messenger that promotes biofilm formation in several bacterial species, but the mechanisms are often unclear. Here, we report that c-di-GMP promotes biofilm formation in mycobacteria in a manner dependent on the ...

Cellular signaling with nitric oxide and cyclic GMP
During the past two decades, nitric oxide signaling has been one of the most rapidly growing areas in biology. This simple free radical gas can regulate an ever growing list of biological processes.

ABSTRACT Acute liver injury (ALI), driven by diverse insults such as drug toxicity and ischemia‐reperfusion, poses a high mortality risk and lacks targeted therapies. While reactive oxygen species (ROS), neutrophil extracellular traps (NETs), and a coordinated cell death pathway PANoptosis have been implicated, their interplay as a unified pathogenic ...

The signaling nucleotide cyclic diguanylate (c-di-GMP) regulates the transition between motile and sessile growth in a wide range of bacteria. Understanding how microbes control c-di-GMP metabolism to activate specific pathways is complicated by the ...

This study proposes a flexible MnO2‐P‐ICG nanofiber patch for laparoscopic treatment of liver tumors. The patch alleviates tumor hypoxia and induces photodynamic therapy (PDT) triggered immunogenic cell death, thereby activating the cGAS‐STING pathway and, in synergy with pachymaran, promoting NK cell–mediated innate immunity. This synergistic strategy

Combination chemotherapy is a common clinical practice in cancer treatment. Here, cyclic RGD (arginylglycylaspartic acid) peptide was introduced to the surface of lipid/calcium/phosphate (LCP) asymmetric lipid layer nanoparticles for the co-delivery of ...

This work identifies M1 macrophage‐mediated inflammation as a key driver of BMSC senescence and bone microstructural deterioration at the enthesis. A tissue‐adhesive hydrogel delivering senomorphic small extracellular vesicles suppresses this inflammatory‐senescent cascade and enhances BMSC resilience by inhibiting the cGAS‐STING‐NF‐κB pathway.

PAIR: Reconstructing Single‐Cell Open‐Chromatin Landscapes for Transcription Factor Regulome Mapping
scATAC‐seq analysis is often constrained by limited sequencing depth, extreme sparsity, and pervasive technical missingness. PAIR is a probabilistic framework that restores scATAC‐seq accessibility profiles by directly modeling the native cell–peak bipartite structure of chromatin accessibility.
