AbstractBackgroundMutations in cyp51A gene are known as main mechanisms of azole resistance in Aspergillus fumigatus, whereas azole‐susceptible strains also carry cyp51A mutations (polymorphisms). The polymorphisms found in Europe mainly consist of two combinations of mutations, that is combinations of five single‐nucleotide polymorphisms (SNPs) of ...

Multiple Aspergillus fumigatus isolates from a patient with two aspergillomas complicating chronic pulmonary aspergillosis were pan-azole resistant. Microsatellite typing was identical for all isolates despite major phenotypic and some growth rate differences. Three different cyp51A mutations were found (G138C, Y431C, and G434C), of which the first two

Azole-resistant Aspergillus fumigatus with TR34/L98H mutations in the cyp51A gene in Iran
Journal de Mycologie Médicale, 2015Introduction Triazole antifungal drugs such as itraconazole, posaconazole and voriconazole are the mainstay of therapy in the management and prophylaxis of aspergillosis. However, in recent years, triazole-resistant clinical isolates of Aspergillus fumigatus have emerged in Europe (the Netherlands, Denmark, Spain, UK, Belgium, Germany and France) and ...

We selected voriconazole-resistant (VCZ-R) Aspergillus fumigatus in the laboratory, characterized the cyp51A gene for possible mutations and evaluated the in vitro activities of voriconazole and anidulafungin alone and in combination against VCZ-R isolates of A.

Multi-azole resistant Aspergillus fumigatus harboring Cyp51A TR46/Y121F/T289A isolated in Japan
Journal of Infection and Chemotherapy, 2016Multi-azole resistant Aspergillus fumigatus carrying TR46/Y121F/T289A was isolated from a patient in Japan in Dec 2013. This strain grouped into the same clade of the ones which were clinically isolated in France and Germany. A. fumigatus harboring this mutation could be rapidly diffused outside the Eurasian continent.

Invasive pulmonary aspergillosis continues to be associated with a high mortality despite timely and appropriate therapy. Although host immunity plays a major role in poor clinical response, antifungal drug resistance cannot be ignored. Our studies were aimed 1) to study the mechanism of drug resistance in voriconazole strains of Aspergillus fumigatus,

Aspergillus fumigatus is the most common etiologic agent of primarily all clinical manifestations of aspergillosis. A steady increase in the number of azole resistant A. fumigatus (ARAF) isolates from environment and clinical samples leading to therapeutic failures in clinical settings have alarmed the mycologists and clinicians worldwide.

Aspergillosis due to Aspergillus fumigatus (A. fum.) is a common and life-threatening infection in immunocompromised patients. A. fum. is increasingly reported to be resistant to azoles essentially in prophylaxis and/or therapy of aspergillosis. Resistance is often associated with alterations in[for full text, please go to the a.m. URL]

ABSTRACTBackgroundAzoles target Cyp51A and Cyp51B in Aspergillus fumigatus. Mutations in cyp51A are known as the primary mechanisms of azole resistance. However, not all of them cause azole resistance. Among them, mutations related to improved susceptibility have not been reported so far.
