Using the model of isolated acini the effect of cytochalasin D (CD) on rat pancreatic secretion in vitro was studied. The influence of CD (0.01-10 micrograms/ml = 0.02-19.7 microM) on amylase secretion and 3H-pulse-labelled protein release was measured under two sets of conditions: (a) basal, and (b) stimulated by 77pM caerulein.

Summary Muscle F-actin diluted to 0.5 mM MgCl2 undergoes a decrease in viscosity which is increased in both rate and extent in the presence of cytochalasin D (CD). At the viscosity plateau, actin with CD hydrolyses ATP at an increased rate dependent on the CD concentration but activates myosin ATPase essentially the same as control actin.

AbstractPregnant rats received intraperitoneal injections of cytochalasin D (CD) on gestational days 7–11. Doses of 400 μg/kg were only minimally and nonsig‐nificantly teratogenic, leading to 2 exencephalic fetuses among the 111 fetuses delivered.

ABSTRACTCharacterization of a cytochalasin D‐resistant mutant of the human parasite Entamoeba histolytica capable of growing at 10 μM cytochalasin is described. The mutant cells also show resistance to 5 mM colchicine and 100 μM cytochalasin B, drugs proved deleterious for wild type trophozoites.

Nocodazole and cytochalasin D induce tetraploidy in mammalian cells

Nocodazole, a rapidly reversible inhibitor of microtubule assembly is useful for preparing mammalian cells synchronized at all stages of mitosis. When synchronized cells are allowed to progress through mitosis in the presence of cytochalasin D, the cleavage furrow is inhibited and dikaryon cells are formed.
