Insights into pegRNA design from editing of the cardiomyopathy‐associated phospholamban R14del mutation

This study reveals how prime editing guide RNA (pegRNA) secondary structure and reverse transcriptase template length affect prime editing efficiency in correcting the phospholamban R14del cardiomyopathy‐associated mutation. Insights support the design of structurally optimized enhanced pegRNAs for precise gene therapy.

C‐mannosylation promotes ADAMTS1 activation and secretion in human testicular germ cell tumor NEC8 cells

C‐mannosylation is a unique form of protein glycosylation. In this study, we demonstrated that ADAMTS1 is C‐mannosylated at Trp562 and Trp565 in human testicular germ cell tumor NEC8 cells. Engineering CRISPR System‐Based Bacterial Outer Membrane Vesicle Potentiates T Cell Immunity for Enhanced Cancer Immunotherapy

This study develops a multifunctional CRISPR‐dCas9‐based OMV platform termed OMV‐C9I12, which facilitates the coexpression of CXCL9 and IL‐12 within tumor cells. This platform enhances T cell recruitment and activation, synergizes with anti‐PD‐1/PD‐L1 immunotherapy, and amplifies antitumor T cell immunity.
