Combining antibody conjugates with cytotoxic and immune‐stimulating payloads maximizes anti‐cancer activity

open access: yesMolecular Oncology, EarlyView.
Methods to improve antibody–drug conjugate (ADC) treatment durability in cancer therapy are needed. We utilized ADCs and immune‐stimulating antibody conjugates (ISACs), which are made from two non‐competitive antibodies, to enhance the entry of toxic payloads into cancer cells and deliver immunostimulatory agents into immune cells.

Specific antitumor activity of anti-CA125 CAR-T lymphocytes against CA125-positive and CA125-negative cells

open access: yesСеченовский вестник
Aim. To evaluate the antitumor efficacy of our developed drug based on cytotoxic T lymphocytes genetically modified with a chimeric antigen receptor (CAR) specific to the CA125 antigen in relation to both CA125-positive and CA125negative cell cultures ...

Hantavirus Inhibits TRAIL-Mediated Killing of Infected Cells by Downregulating Death Receptor 5

open access: yesCell Reports, 2019
Summary: Cytotoxic lymphocytes normally kill virus-infected cells by apoptosis induction. Cytotoxic granule-dependent apoptosis induction engages the intrinsic apoptosis pathway, whereas death receptor (DR)-dependent apoptosis triggers the extrinsic ...

Spatiotemporal dynamics of a glioma immune interaction model

open access: yesScientific Reports, 2021
We report a mathematical model which depicts the spatiotemporal dynamics of glioma cells, macrophages, cytotoxic-T-lymphocytes, immuno-suppressive cytokine TGF-β and immuno-stimulatory cytokine IFN-γ through a system of five coupled reaction-diffusion ...

Regional peculiarities of specific immunity in inhabitants of different mountain heights of the Kyrgyz Republic

open access: yesMedicine Science, 2023
Kyrgyzstan's challenging natural conditions and fragile mountain ecosystems can weaken the immune system of individuals residing permanently in high-altitude areas.

The PI3Kδ inhibitor roginolisib (IOA‐244) preserves T‐cell function and activity

open access: yesMolecular Oncology, EarlyView.
Identification of novel PI3K inhibitors with limited immune‐related adverse effects is highly sought after. We found that roginolisib and idelalisib inhibit chronic lymphocytic leukemia (CLL) cells and Treg suppressive functions to similar extents, but roginolisib affects cytotoxic T‐cell function and promotion of pro‐inflammatory T helper subsets to a

Cotargeting TREM2 and IL2 pathways triggers multipronged anticancer immunity

open access: yesMolecular Oncology, EarlyView.
Von Locquenghien et al. report that MiTE‐144, a triggering receptor expressed on myeloid cells 2 (TREM2) blocking antibody fused to interleukin‐2 (IL2) variant with tumour microenvironment restricted activation, demonstrates superior anticancer efficiency in a preclinical setting.

Adoptive cancer immunotherapy using DNA-demethylated T helper cells as antigen-presenting cells

open access: yesNature Communications, 2018
A critical determinant of tumor eradication by adoptive immunotherapy is the tumor associated antigen recognized by cytotoxic T lymphocytes. Here the authors generate ex vivo autologous cytotoxic T lymphocytes by exposure to antigens induced by DNA ...
