Study of the multiple incorporation of modified nucleotides into the growing DNA strand

open access: yesТонкие химические технологии, 2021
Objectives. This study investigated the substrate properties of the modified derivatives of triphosphates of purine and pyrimidine deoxynucleosides (5-propynyl-2’-deoxyuridine-5’-triphosphate, 5-propynyl2’-deoxycytidine-5’-triphosphate, 5-methyl-2 ...

ATR inhibition facilitates targeting of leukemia dependence on convergent nucleotide biosynthetic pathways. [PDF]

open access: yes, 2017
Leukemia cells rely on two nucleotide biosynthetic pathways, de novo and salvage, to produce dNTPs for DNA replication. Here, using metabolomic, proteomic, and phosphoproteomic approaches, we show that inhibition of the replication stress sensing kinase ...

Kaiso interacts with p120-catenin to regulate β-catenin expression at the transcriptional level. [PDF]

open access: yesPLoS ONE, 2014
BACKGROUND: We have reported that p120-catenin could regulate β-catenin transcription in lung cancer cells, but the specific mechanism is unclear. METHODS AND RESULTS: In this study, bisulfite sequencing PCR showed that the β-catenin promoter region in ...

Activation-induced deoxycytidine deaminase (AID) co-transcriptional scanning at single-molecule resolution [PDF]

open access: yes, 2015
Activation-induced deoxycytidine deaminase (AID) generates antibody diversity in B cells by initiating somatic hypermutation (SHM) and class-switch recombination (CSR) during transcription of immunoglobulin variable (IgV) and switch region (IgS) DNA ...

Demethylation by low-dose 5-aza-2′-deoxycytidine impairs 3D melanoma invasion partially through miR-199a-3p expression revealing the role of this miR in melanoma

open access: yesClinical Epigenetics, 2019
Background Efficient treatments against metastatic melanoma dissemination are still lacking. Here, we report that low-cytotoxic concentrations of 5-aza-2′-deoxycytidine, a DNA demethylating agent, prevent in vitro 3D invasiveness of metastatic melanoma ...

Purine nucleoside phosphorylase enables dual metabolic checkpoints that prevent T cell immunodeficiency and TLR7-associated autoimmunity

open access: yesThe Journal of Clinical Investigation, 2022
Purine nucleoside phosphorylase (PNP) enables the breakdown and recycling of guanine nucleosides. PNP insufficiency in humans is paradoxically associated with both immunodeficiency and autoimmunity, but the mechanistic basis for these outcomes is ...

LC-MS-MS quantitative analysis reveals the association between FTO and DNA methylation. [PDF]

open access: yesPLoS ONE, 2017
Fat mass and obesity-associated protein (FTO) is α-ketoglutarate-dependent dioxygenase and responsible for demethylating N6-methyladenosine (m6A) in mRNA, 3-methylthymine (m3T) in single-stranded DNA (ssDNA) and 3-methyluracil (m3U) in single-stranded ...

Gender effects on cytidine analogue metabolism and myelodysplastic syndrome treatment outcomes [PDF]

open access: yes, 2012
In vivo, half-lives of cytidine analogues such as 5-azacytidine and decitabine, used to treat myelodysplastic syndromes (MDS), are determined largely by cytidine deaminase (CDA), an enzyme that rapidly metabolizes these drugs into inactive uridine ...

Increased levels of DNA methyltransferases are associated with the tumorigenic capacity of prostate cancer cells [PDF]

open access: yes, 2012
DNA methylation might be the earliest somatic genome changes in prostate cancer that also play an important role in the process of tumor invasion, growth and metastasis.

IMPDH inhibition enhances cytarabine efficacy in SAMHD1‐expressing leukaemia cells via guanine nucleotide depletion

open access: yesMolecular Oncology, EarlyView.
Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.
