Organoids in pediatric cancer research

open access: yesFEBS Letters, EarlyView.
Organoid technology has revolutionized cancer research, yet its application in pediatric oncology remains limited. Recent advances have enabled the development of pediatric tumor organoids, offering new insights into disease biology, treatment response, and interactions with the tumor microenvironment.

A Direct Search Algorithm for Global Optimization

open access: yesAlgorithms, 2016
A direct search algorithm is proposed for minimizing an arbitrary real valued function. The algorithm uses a new function transformation and three simplex-based operations.

Phosphatidylinositol 4‐kinase as a target of pathogens—friend or foe?

open access: yesFEBS Letters, EarlyView.
This graphical summary illustrates the roles of phosphatidylinositol 4‐kinases (PI4Ks). PI4Ks regulate key cellular processes and can be hijacked by pathogens, such as viruses, bacteria and parasites, to support their intracellular replication. Their dual role as essential host enzymes and pathogen cofactors makes them promising drug targets.

Protein pyrophosphorylation by inositol pyrophosphates — detection, function, and regulation

open access: yesFEBS Letters, EarlyView.
Protein pyrophosphorylation is an unusual signaling mechanism that was discovered two decades ago. It can be driven by inositol pyrophosphate messengers and influences various cellular processes. Herein, we summarize the research progress and challenges of this field, covering pathways found to be regulated by this posttranslational modification as ...

A Hybrid Direct Search and Model-Based Derivative-Free Optimization Method with Dynamic Decision Processing and Application in Solid-Tank Design

open access: yesAlgorithms, 2023
A derivative-free optimization (DFO) method is an optimization method that does not make use of derivative information in order to find the optimal solution. It is advantageous for solving real-world problems in which the only information available about

Geometry of interpolation sets in derivative free optimization [PDF]

open access: yesMathematical Programming, 2006
The aim of the paper is to describe derivative free optimization methods and to prove their convergence. To compute new iterates in those methods a polynomial interpolation model of the objective function is minimized. An essential tool for proving convergence of those algorithms is an error bound between the interpolation model and the objective ...

Structural instability impairs function of the UDP‐xylose synthase 1 Ile181Asn variant associated with short‐stature genetic syndrome in humans

open access: yesFEBS Letters, EarlyView.
The Ile181Asn variant of human UDP‐xylose synthase (hUXS1), associated with a short‐stature genetic syndrome, has previously been reported as inactive. Our findings demonstrate that Ile181Asn‐hUXS1 retains catalytic activity similar to the wild‐type but exhibits reduced stability, a looser oligomeric state, and an increased tendency to precipitate ...
