Background and Purpose Liver X receptors (LXRs) are promising therapeutic targets for alleviating Alzheimer's disease (AD) symptoms. We assessed the impact of the semi‐synthetic LXR agonist 22‐ketositosterol on disease progression in an AD mouse model.

Signaling regulates activity of DHCR24, the final enzyme in cholesterol synthesis[S]
The role of signaling in regulating cholesterol homeostasis is gradually becoming more widely recognized. Here, we explored how kinases and phosphorylation sites regulate the activity of the enzyme involved in the final step of cholesterol synthesis, 3β ...

Cholesterol Within Phosphatidylcholine Liposomes Attenuates the Expression of Cytokines
We previously showed that liposomes made of phosphatidylcholine, a natural phospholipid found in plasma membranes, exosomes, and lipid nanoparticles, strongly induce TNF‐α expression in macrophages. Here, we found that adding free cholesterol to these liposomes significantly reduces this response by inhibiting NF‐κB activation.

Macrophage-derived upd3 Cytokine causes impaired glucose homeostasis and reduced lifespan in drosophila fed a lipid-rich diet [PDF]
Long-term consumption of fatty foods is associated with obesity, macrophage activation and inflammation, metabolic imbalance, and a reduced lifespan. We took advantage of Drosophila genetics to investigate the role of macrophages and the pathway(s) that ...

Host Organelle Interactions Facilitate Cholesterol Acquisition by Trypanosoma cruzi Amastigotes
ABSTRACT Chagas disease, caused by the protozoan Trypanosoma cruzi, is a major neglected disease in Latin America. The amastigote, the replicative intracellular form, is essential for infection persistence in vertebrate hosts. These forms exhibit remarkable adaptability, modulating metabolism and growth according to host cell resource availability ...

Abstract Compared with 2D monolayers, 3D models more closely mimic native muscle tissue and allow functional measurements. A more complete understanding of how culture conditions and duration affect myotube maturity/function is crucial for validating the transition to 3D systems.

The use of the Dhcr7 knockout mouse to accurately determine the origin of fetal sterols
Mice with a targeted mutation of 3β-hydroxysterol Δ7-reductase (Dhcr7) that cannot convert 7-dehydrocholesterol to cholesterol were used to identify the origin of fetal sterols.

A review of the putative antiseizure and antiepileptogenic mechanisms of action for soticlestat
Abstract Soticlestat (TAK‐935) is a potent and selective inhibitor of cholesterol 24‐hydroxylase (CYP46A1), an enzyme primarily expressed in the brain that catabolizes cholesterol to 24S‐hydroxycholesterol (24HC). In the ELEKTRA phase II clinical trial, soticlestat reduced seizure frequency in patients with developmental and epileptic encephalopathies,

Lipid peroxidation biomarkers for evaluating oxidative stress in equine neuroaxonal dystrophy. [PDF]
BackgroundEquine neuroaxonal dystrophy/equine degenerative myeloencephalopathy (eNAD/EDM) is a neurodegenerative disorder affecting genetically predisposed foals maintained on an α-tocopherol (α-TOH) deficient diet.
