Quantitative Profiling of Nanoscopic Protein Aggregates Reveals Specific Fingerprint of TDP‐43‐Positive Assemblies in Motor Neuron Disease

open access: yesAdvanced Science, EarlyView.
Single‐molecule fluorescence microscopy is combined with proteomic profiling to fingerprint the morphology and composition of TDP‐43 protein aggregates found in post‐mortem motor neuron disease brain tissues. Aggregate fingerprints can distinguish disease from control donors and detect unexpected TDP‐43 pathology in SOD1‐MND.

Pathogenicity of Mediator Complex Subunit 27 (MED27) in a Neurodevelopmental Disorder with Cerebellar Atrophy

open access: yesAdvanced Science, EarlyView.
MED27 is one of the 26 subunits in the human Mediator complex (MED). Neurodevelopmental disorder‐causing MED27 genetic variants induce instability of MED, leading to disrupted DNA occupancy, altered chromatin interaction, and subsequent transcriptional dysregulation of critical downstream genes, including master regulatory transcription factors ...

Human Brain Cell‐Type‐Specific Aging Clocks Based on Single‐Nuclei Transcriptomics

open access: yesAdvanced Science, EarlyView.
Muralidharan and colleagues develop cell‐type‐specific transcriptomic aging clocks using single‐nucleus RNA sequencing of human post mortem prefrontal cortex samples. These clocks accurately predict age and identify distinct aging trajectories in specific brain cell types.

Contribution of Prostaglandin E2‐Induced Neuronal Excitation to Drug Resistance in Glioblastoma Countered by a Novel Blood–Brain Barrier Crossing Celecoxib Derivative

open access: yesAdvanced Science, EarlyView.
Glioblastoma (GBM) is an aggressive brain cancer that often recurs and resists chemotherapy. This study found that a chemical called PGE2 causes surrounding brain cells (neurons) to become over‐excited, which makes GBM cells resistant to treatment. Researchers developed a new drug, “Compound 11,” which can cross into the brain and effectively block ...

Neuro‐Cancer Interactions Shape Glioma Intratumoral Heterogeneity

open access: yesAdvanced Science, EarlyView.
This review proposes a conceptual framework for understanding glioma intratumoral heterogeneity shaped by neuro‐cancer interactions: functional patterns and transcriptional states. Functional patterns manifest as three major neural‐like adaptations with spatial and potentially hierarchical relationships, including neurogliomal synapses, glioma networks,
