Structural Characterization of Insoluble Dextran Produced by Leuconostoc mesenteroides NRRL B-1149 in the Presence of Maltose

Leuconostoc mesenteroides NRRL B-1149 is known to produce dextran with 52 % α-(1→6) and 40 % α-(1→3) linkages. Low solubility of dextran in water is associated with the presence of high percentage of α-(1→3) linkages.

Studies on Dextransucrase Part I

(1) Leucanostoc mesenteroides IAM 1046により,デキストランシュクラーゼを生産させるための培地,および培養条件を検討した. (2)上記菌株のデキストランシュクラーゼの部分的精製を行ない,硫安塩析およびDEAE-セルロースによるクロマトグラフィーを期いて,比活性が培養液の24~56倍の精製酵素を得た. (3)部分精製したデキストランシュクラーゼの酵素化学的性質を検討した.その結果,この酵素は作用最適pH 5.0,最適温度30°C, pH 4.0~7.5において安定であるが, 40°C以上にて速やかに失活するなどのことがわかった.またMichaelis恒数は30°C, pH 5.0で1.57×10-2 (M)であった.
