Large‐Diameter DNA‐Scaffolded Nanopores Enabled by Loosely Packed Peptides for Single‐Molecule Sensing

A nanopore composed of transmembrane peptides and a DNA scaffold that assists peptide assembly was constructed. By employing peptides containing the unnatural amino acid 2‐aminoisobutyric acid (Aib), which promotes loose packing between helices, a large‐diameter nanopore was achieved.

The nitrosuccinate lyase CreD catalyzes C–NO2 bond formation using nitrite in water and shows synthetic practicality with high turnover numbers up to 102,000.
