Modifying effects of age, sex, and mouse strain on diethylnitrosamine (DEN) carcinogenesis have been investigated in C57BL/6Jx C3HeB/FeJ F1 (B6C3F1) and C3HeB/FeJxA/J F1 (C3AF1) hybrid mice. Animals each received four IP injections of 1.5 or 3.0 micrograms DEN/g body weight.

Adrenergic component in the hepatotropic carcinogenic effect of diethylnitrosamine

The effect of norepinephine, an adrenomietic drug, and of pyrroxane, its antagonist, on diethylnitrosoamine (DENA) hepatocarcinogenesis was studied in albino rats. Norepinephrine was found to stimulate carcinogenesis, whereas pyrroxane--to inhibit this process; the latter drug decreased the incidence of multicentric tumours of the liver.

Formation of O6-ethylguanine (O6EG) in trachea DNA was measured 12 h after acute i.p. administration of 50, 100 and 200 mg diethylnitrosamine (DEN)/kg body wt to Syrian golden hamsters. Purine bases were fractionated by h.p.l.c., and ethylated guanines were quantified optically by fluorescence spectrophotometry.

The dialkyl nitrosamines, of which dimethylnitrosamine and diethylnitrosamine are good examples, belong to the larger class of N-nitroso compounds, many of which are potent carcinogens in a large variety of animal species. In 1954 Barnes and Magee (1) demonstrated that dimethylnitrosamine was acutely hepatotoxic to laboratory animals.

Effects of diethylnitrosamine and cigarette smoke on hamsters.

One hundred two 2-month-old male Syrian hamsters received 12 weekly sc injections of 0.25 mg diethylnitrosamine (DEN). Half the animals were also exposed for 10 minutes to cigarette smoke in Hamburg II-type smoking machines, three times per day, 5 days per week, for their life-span. The other half received sham exposure.

Kinetics of hepatocarcinogenesis was evaluated in 15-day-old male C57BL/6J X C3HeB/FeJ F1 mice using a nontoxic carcinogenic dose range of diethylnitrosamine (DEN). The carcinogen was injected i.p. once, and the animals were killed according to the protocol. Two studies were carried out sequentially over a period of 4 years.
