The effect of norepinephine, an adrenomietic drug, and of pyrroxane, its antagonist, on diethylnitrosoamine (DENA) hepatocarcinogenesis was studied in albino rats. Norepinephrine was found to stimulate carcinogenesis, whereas pyrroxane--to inhibit this process; the latter drug decreased the incidence of multicentric tumours of the liver.

Formation of O6-ethylguanine (O6EG) in trachea DNA was measured 12 h after acute i.p. administration of 50, 100 and 200 mg diethylnitrosamine (DEN)/kg body wt to Syrian golden hamsters. Purine bases were fractionated by h.p.l.c., and ethylated guanines were quantified optically by fluorescence spectrophotometry.

The dialkyl nitrosamines, of which dimethylnitrosamine and diethylnitrosamine are good examples, belong to the larger class of N-nitroso compounds, many of which are potent carcinogens in a large variety of animal species. In 1954 Barnes and Magee (1) demonstrated that dimethylnitrosamine was acutely hepatotoxic to laboratory animals.
