Liquid biopsy epigenetics: establishing a molecular profile based on cell‐free DNA
Cell‐free DNA (cfDNA) fragments in plasma from cancer patients carry epigenetic signatures reflecting their cells of origin. These epigenetic features include DNA methylation, nucleosome modifications, and variations in fragmentation. This review describes the biological properties of each feature and explores optimal strategies for harnessing cfDNA ...

Fast Distributed PageRank Computation
Over the last decade, PageRank has gained importance in a wide range of applications and domains, ever since it first proved to be effective in determining node importance in large graphs (and was a pioneering idea behind Google's search engine).

Clinical trials on PARP inhibitors in urothelial carcinoma (UC) showed limited efficacy and a lack of predictive biomarkers. We propose SLFN5, SLFN11, and OAS1 as UC‐specific response predictors. We suggest Talazoparib as the better PARP inhibitor for UC than Olaparib.

Asynchronous Distributed Semi-Stochastic Gradient Optimization
With the recent proliferation of large-scale learning problems, there have been a lot of interest on distributed machine learning algorithms, particularly those that are based on stochastic gradient descent (SGD) and its variants.

HDAC4 is degraded by the E3 ligase FBXW7. In colorectal cancer, FBXW7 mutations prevent HDAC4 degradation, leading to oxaliplatin resistance. Forced degradation of HDAC4 using a PROTAC compound restores drug sensitivity by resetting the super‐enhancer landscape, reprogramming the epigenetic state of FBXW7‐mutated cells to resemble oxaliplatin ...

Two‐stage personalized federated learning based on sparse pretraining
Aiming at solving the performance degradation of federated learning (FL) under heterogeneous data distribution, personalized FL (PFL) was proposed. It is designed to produce a dedicated model for each client.

We developed and validated a DNA methylation–based biomarker panel to distinguish pleural mesothelioma from other pleural conditions. Using the IMPRESS technology, we translated this panel into a clinically applicable assay. The resulting two classifier models demonstrated excellent performance, achieving high AUC values and strong diagnostic accuracy.
